黄瓜霜霉病菌不同发育阶段对烯肟菌酯的敏感性

摘  要:
【目的】研究黄瓜霜霉病菌不同发育阶段对烯肟菌酯的敏感性,明确建立敏感性基线的生物测定方法。【方法】本研究以专性寄生菌黄瓜霜霉病菌为靶标菌,采用凹玻片法和叶碟保湿法,测定了该病原菌不同发育阶段对烯肟菌酯的敏感性。【结果】研究结果表明,烯肟菌酯对黄瓜霜霉病菌休止孢的萌发没有影响,对黄瓜霜霉病菌游动孢子释放、游动孢子游动、芽管伸长、菌丝扩展、孢囊梗形成及孢子囊产生均有抑制作用,而且对黄瓜霜霉病菌的游动孢子释放、游动孢子游动和芽管伸长有显著的抑制作用。其中对黄瓜霜霉病菌的游动孢子释放和芽管伸长EC50值分别为0.0250和0.0037μg·ml-1;并且当烯肟菌酯浓度为0.05μg·ml-1时能使游动孢子消解,抑制休止孢的形成;当药剂浓度为5μg·ml-1时,对病斑的扩展和孢子囊产生抑制率分别达到60.41%和92.10%,且对孢囊梗的产生也有显著的抑制作用。【结论】综合考虑试验方法的可操作性和稳定、准确性,推荐使用叶碟保湿法测定烯肟菌酯对菌丝扩展的EC50,用以建立敏感基线并进行田间抗药性监测。

摘  要:
【Objective】 The bioassay method of baseline sensitivity was developed on the basis of the effect of enostrobilurin on Pseudoperonospora cubensis at different development stages during its life cycle. 【Method】 The sensitivity was determined by concave-slide and the leaf discs assay in vitro respectively. 【Result】The results showed that enostrobilurin did not show any effect on cystospore germination, but prevented the zoospore release, zoospore motility, germ tube elongation, hyphal growth, sporangiophore emergence from stomata and zoosporangial formation. The EC50 values of enostrobilurin to zoospore release and germ tube elongation were 0.0250 μg·ml-1, 0.0037 μg·ml-1 respectively. Enostrobilurin resulted in zoospore breakdown at 0.05 μg·ml-1 and inhibited obviously hyphal growth, sporangiophore emergence from stomata and sporangial formation at 5 μg·ml-1. 【Conclusion】 These studies suggested that leaf discs assay in vitro was an accurate and reliable method to determine the EC50 values of flumorph to P.cubensis, the baseline sensitivity and the resistance strain in field.
