五条鰤源美人鱼发光杆菌杀鱼亚种的分离鉴定及药物敏感试验

摘  要:
2007年5月广东汕尾红海湾网箱养殖的五条鰤爆发主要症状为脾脏、肾脏出现许多灰白色小结节的疾病。从病鱼体内分离出1株优势菌株,命名为SWZX5,为革兰氏阴性杆菌。SWZX5对剑尾鱼有强致病性,其半数致死量为5.8×105CFU/g。综合细菌在形态、生理生化特性及16S rRNA系统发育树等方面的研究结果,确认分离菌株SWZX5属于美人鱼发光杆菌杀鱼亚种(Photobacterium damselae subsp.piscicida)。15种中草药提取液的抑菌和杀菌试验显示,五倍子的功效最好,MIC和MBC均为0.05mg/mL;其次是黄连、石榴皮,两者MIC均为0.20 mg/mL,MBC均为0.40 mg/mL;黄芪功效较差。20种化学抗菌类药物的抑菌试验显示,分离菌株SWZX5对供试的氨曲南、恩诺沙星、左旋氧氟沙星、头孢呋辛钠等16种抗菌药物敏感;对卡那霉素、苯唑西林等4种抗菌药物中度敏感或耐药。

摘  要:
An epizooty occurred among the net-cage cultured yellowtail(Seriola quinqueradiata) in Shanwei,Guangdong province,in 2007.The main symptom of diseased fish was the appearance of many gray nodules in the spleen and kidney.A Gram-negative,rod shaped bacterium(designated strain SWZX5) was isolated from the spleen of diseased fish.Artificial infection proved that the isolate was virulent to swordtail(Xiphophorus helleri) with the LD50 being calculated as 5.8 ×105 CFU/g.A phylogenetic tree was constructed by comparing the 16S rRNA gene sequence of the isolate with other relative bacteria species in the RDP and GenBank databases.In the phylogenetic tree,the isolates and some Photobacterium damselae subsp.piscicida strains converge and constitute a branch,which confirms the identication of the isolate as P.damselae subsp.piscicida.According to the morphological,physiological,biochemical characteristics and 16S rRNA gene analysis,the isolate was identified as Ph.damselae subsp.piscicida.The minimum inhibitory concentrations(MIC) and minimum bactericidal concentrations(MBC)of 15 kinds of Chinese herbal medicines extracted by decoction by boiled water to the isolates were determined.It was found that there were best bacteriostatic and bactericidal effect of Galla Chinensis,followed by Rhizoma Coptidis and Pericarpium Granati.The MIC and MBC were found to be 0.05 mg/mL for Galla Chinensis,0.2 0mg/ mL and 0.40 mg /mL for Rhizoma Coptidis,0.20 mg /mL and 0.40 mg/mL for Pericarpium Granati.Antibiotic sensitivity test of 20 kinds of chemical antimicrobial drugs showed the isolate was sensitive to 16 kinds of antibiotics including Aztreonam(30 μg),Enrofloxacine(5 μg),Cefuroxime Sodium(30 μg),Levofloxacin(5 μg).
