猪源乳酸杆菌对致病性沙门菌、大肠埃希菌黏附仔猪小肠上皮细胞的影响

摘  要:
为了通过仔猪小肠上皮细胞(ZYM-SIEC-02)体外培养模型,观察猪源乳酸杆菌对细胞的黏附作用,以及对致病性沙门菌、大肠埃希菌黏附细胞的竞争、排斥和置换作用。采用乳酸杆菌在体外与ZYM-SIEC-02共同孵育镜下观察其黏附效果;乳酸杆菌与致病性沙门菌、大肠埃希菌共同黏附ZYM-SIEC-02观察其黏附竞争性;用乳酸杆菌预处理ZYM-SIEC-02后再加入致病性沙门菌、大肠埃希菌孵育观察其黏附排斥性;用致病性沙门菌、大肠埃希菌预处理ZYM-SIEC-02后再加入乳酸杆菌孵育观察其黏附置换性。结果表明:乳酸杆菌可以黏附ZYM-SIEC-02,具有剂量效应。乳酸杆菌与致病性沙门菌或大肠埃希菌同时加入细胞一起培养时,能竞争性的黏附细胞且黏附抑制率也具有剂量效应。乳酸杆菌预处理细胞后再加入致病菌,高含量乳酸杆菌(1010 mL-1)能使致病菌的黏附率下降。但是,用致病菌预处理细胞后再加入乳酸杆菌,只有高含量的乳酸杆菌可以置换致病菌,而低含量乳酸杆菌却能增加致病菌对细胞的黏附。研究结果为乳酸菌作为微生态制剂用于临床上防治仔猪沙门菌病、大肠埃希菌病,减少食物源性污染和提高食品安全提供试验依据和理论基础。

摘  要:
To evaluate the adhesion of pig Lactobacillus strains on Pig Small Intestinal Epithelial Cell line(ZYM-SIEC-02),the competition,exclusion and displacement of adhesion to Salmonella,Escherichia coli on ZYM-SIEC-02 cells were investigated.The adhesion effect was observed when Lactobacillus were co-cultivated with ZYM-SIEC-02 cells;the adhesion competive was observed when Lactobacillus were co-cultivated with Salmonella,Escherichia coli and ZYM-SIEC-02 cells;after the ZYMSIEC-02 were pretreated by Lactobacillus,the adhesion exclusion was observed when Salmonella,Escherichia coli were co-cultivated with ZYM-SIEC-02 cells;after the ZYM-SIEC-02 were pretreated by Salmonellaand Escherichia coli,the adhesion displacement was observed when Lactobacillus were co-cultivated with ZYM-SIEC-02 cells.The results show that the Lactobacillus exhibited dose dependent adhesion to ZYM-SIEC-02.The ability to inhibit the competition adhesion of Salmonellaor Escherichia coli appeared to depend on the dose by 1010 mL-1 Lactobacillus.Adhesion percent of Salmonella was reduced by pretreating the cells with 1010 mL-1 Lactobacillus prior to Lactobacillus adhesion to ZYM-SIEC-02 cell monolayers.No displacement was detected at low concentration of Lactobacillus.These results would offered a new efficient way for preventing and theraping pig Salmonellosis,Escherichia coli,reducing food-born pollution and raising foods safety.
