利用DD-PCR技术研究扬稻6号基因的差异表达

摘  要:
利用DD-PCR技术对扬稻6号参与的强优势组合(扬两优6号和两优培九)、明恢63参与的弱优势组合、R13参与的中强优势组合以及亲本的基因表达进行了分析。根据基因在杂交组合和双亲中的表达情况分为8种类型,统计30个引物组合在11个参试材料中的表现,获得314组差异条带,大小在250 bp至1 000 bp之间,其中强优势组合的差异表达带型UNP1、UNF1和ABF1明显低于弱优势组合,而DMP2和MONO则明显高于弱优势组合。用8种类型所占的比例对各参试材料进行聚类分析,结果显示,扬稻6号参与的强优势组合和明恢63参与的弱势组合明显被分为两类,两者在基因表达模型上存在一定差异。此外,在基因表达水平上探讨了扬稻6号杂种优势的特殊分子基础。

摘  要:
Gene expression differences among high-heterosis crosses with Yangdao 6 as male parent,middle-heterosis crosses with R13 as parent and low-heterosis crosses with Minghui 63 as parent were analyzed by differential display PCR(DD-PCR) technique.Eight types of differential expression patterns were classified and 314 specific bands with lengths of 250-1 000 bp were obtained based on gene expression in F1 and their parents using 30 primer combinations.The values of UNP1,UNF1 and ABFl in high-heterosis crosses were lower than those in low-heterosis crosses,but DMP2 and MONO were higher in high-heterosis crosses than those in low-heterosis crosses.High-heterosis crosses derived from Yangdao 6 and low-heterosis crosses from Minghui 63 showed significant differences in gene expression and were clustered into two groups according to the percentage of eight types of differential expression pattern.Molecular mechanism of Yangdao 6 showing high heterosis was also discussed.
