离体条件下睾酮与IGF1对培养的鹿茸增生层细胞分裂繁殖的影响

摘  要:
采取离体培养的方法研究了单独的睾酮或睾酮与IGF1配伍对鹿茸增生层间质细胞或软骨细胞所起的促有丝分裂作用,同时对从组织分离出的细胞有无睾酮特异结合位点进行了分析。结果表明,在这些细胞中存在睾酮的特异结合位点,但无论生理剂量还是较低水平的睾酮都没有直接地促进细胞有丝分裂的作用。睾酮(0.001~1000nmol)与IGF1(0.3~10.0nmol)配伍并没有增强IGF1的促有丝分裂作用,相反,在一定剂量下却减弱了IGF1的促有丝分裂作用。因此,这些结果即不支持低水平睾酮具有促使茸角形成的作用,也不支持睾酮通过致敏茸角细胞从而使IGF1的促有丝分裂更为敏感的假说。

摘  要:
An experiment in vitro was conducted to determine whether testosterone alone or with IGF-I had mitogenic effects on mesenchymal or cartilaginous cells derived from the tip of regenerating antlers. In addition,a binding assay was also carried out to determine whether the specific binding sites for testosterone were preserved after cell disaggregation. Results showed that testosterone either in physiological concentrations or at low levels did not exert direct mitogenic effects on antler cells derived from the proliferation zone in serum-free medium in vitro. Likewise,testosterone in a very wide range of concentrations (0.001~1000nmol) not only failed to enhance,but at certain levels (0.1~5.0nmol) impaired the mitogenic effects of IGF-I on these antler cells in vitro. Therefore,these results do not support a conclusion that low level testosterone has growth-promoting effects on antler formation or the hypothesis that testosterone effects may be achieved through sensitizing these antler cells to the mitogenic effects of IGF-I.
