金线兰愈伤组织诱导的最佳激素配比

摘  要：
以金线兰根状茎、幼茎、叶片、离体胚为外植体,4种激素2,4-D、NAA、6-BA、ZT采用L9(34)正交表处理,对愈伤组织诱导的条件进行研究。结果表明,6-BA和2,4-D在诱导金线兰愈伤组织中作用极为显著,4种激素对愈伤组织的诱导作用大小依次为,6-BA>2,4-D>NAA>ZT,初步断定金线兰愈伤组织诱导的最佳激素配比是0.2 mg/L 2,4-D+0.9 mg/L NAA+1.0 mg/L 6-BA+0.50 mg/L ZT;根状茎和幼茎作为外植体更易诱导愈伤组织形成;暗培养较光暗交替更易诱导愈伤组织生长。

摘  要：
Using the rhizome,leaf blade,steam and in vitro embryo of Anoectochilus roxburghii as explants,the inducing conditions of callus were studied by L9(34) orthogonal design of four kinds of plant growth regulators,2,4-D,6-BA,NAA and ZT.The results showed that 6-BA and 2,4-D played extremely important roles in induction of callus.The order of the efficiency of the four regulators was 6-BA>2,4-D>NAA>ZT.The best medium for calli inducement was MS+0.2 mg/L 2,4-D+0.9 mg/L NAA+1.0 mg/L 6-BA +0.50 mg/L ZT.Inducing callus from rhizome and stem was easier;and callus browning rate under dark was lower than that under light-dark alternate conditions.
