水牛牛巴贝斯虫液氮保存方法的比较

摘  要:
试验分别以二甲基亚砜(DMSO)和聚乙烯吡咯烷酮(PVP)作保护剂,采用一步法(直接投入液氮)和二步法(气相预冷15~30min后投入液氮)液氮保存水牛牛巴贝斯虫。保存不同时间的样品复苏后接种去牌水牛犊,均引发了典型的牛巴贝斯虫病,从结果可以看出:DMSO和PVP都可以用作牛巴贝斯虫液氮保存的保护剂,二步法的冻存效率高于一步法。考虑到毒性作用的因素,保存牛巴贝斯虫时以PVP作保护剂采用二步法冻存是较为理想的办法。

摘  要:
Babesia bovis parasitized in buffalo were cryopreserved with solution of 10% polyvingpyrrolidone (PVP) or 8% dimethyl sulpboxide (DMSO) as cryoprotectant in liquid nitrogen. Samples were cooled by one-step (Samples were directly immersed in liquid N2) or by two-step (Samples were held in gas phase of liquid N2 before immersed in liquid N2). The viability of samples cryopreserved with different time intervals was tested by inoc-ulating splenectomized buffalo calves after thawing.,The results showed that both DMSO and PVP could be used as cryoprotecant for B. bovis parasitized in buffalos, the two-step cooling could improve the survival of parasites. Allowing for the toxicity of DMSO, the au-thors regard that using PVP as cryoprotectant and cooling by two-step is better.
