摘  要：
【目的】探讨牛II型胶原诱导SD大鼠构建类风湿关节炎(Rheumatoid arthritis,RA)模型的可行性,为今后深入研究RA提供理想的动物模型。【方法】选择60只4周龄的SD大鼠,依据体重相近的原则随机分组(模型组50只,对照组10只),模型组通过注射牛Ⅱ型胶原建模,并通过体征观察、病理组织切片评价、钼靶影像学检测等判定建模是否成功。【结果】通过尾静脉注射牛Ⅱ型胶原免疫SD大鼠的建模成功率高达90.0%。与对照组SD大鼠相比,模型组SD大鼠体重减轻,活动减少,精神萎靡,先是后足小趾或踝关节红肿,之后炎症漫延至前肢,最终发展成慢性、多发性、对称性关节炎;模型组SD大鼠关节滑膜层次增多,且有大量炎性细胞浸润,软骨及骨组织受损,关节腔结构紊乱;模型组SD组大鼠从初次免疫第10周开始,其关节间隙狭窄、模糊,骨质疏松,关节软骨及骨组织受损。【结论】SD大鼠是研究RA发病机制及研发抗风湿药物的理想动物模型,可丰富该疾病动物模型的选择。

摘  要：
【Objective 】In order to provide ideal animal model for further research on rheumatoid arthritis(RA), the present experiment was conducted to investigate feasibility of establishing model for SD rat arthritis induced with bovine II-type collagen. 【Method】Sixty 4-week-old SD rats were selected and randomly grouped according to similar weight principle. Fifty of them was assigned to model group, and ten of them was assigned to control group. Then bovine II-type collagen was injected into rats in the model group, so as to establish model for SD rat arthritis. And later the established model was assessed through signs observation, HE pathological evaluation and molybdenum target imaging inspection, in order to detect whether the model establishment was successful or not. 【Result】The results showed that the success rate of establishing model for SD rat arthritis induced with bovine II-type collagen reached up to 90.0%. Compared with SD rats of control group, the SD rats of model group showed obvious symptoms, such as weight loss, activity decrease, drooping spirit, firstly little toe or ankle joint of metapodium becoming red and swollen, then inflammation spreading to fore limb,eventually developing into chronic and symmetric polyarthritis. In addition, there were more synovial membrane layers, a large number of inflammatory cells infiltration, destructed cartilage and bone, disordered structure of articular cavity to be found in SD rats of model group. Furthermore, the narrowed vague joint space, osteoporosis, damaged articular cartilage and bone could be found in the SD rats of model group from the 10 thweek after primary immune. 【Conclusion】SD rat is the ideal animal model for research on RA pathogenesis and anti-rheumatoid drugs, is a new animal model for RA.
