猪伪狂犬病毒变异株与大肠杆菌、志贺氏菌混合感染的病原鉴定及分析

摘  要:
【目的】确诊广西金陵某种猪场2014年发病的原因,了解猪伪狂犬病毒(PRV)变异株与细菌混合感染的趋势,为有效防控PR疫情发展与制定防控措施提供参考依据。【方法】以常规的实验室检测方法进行细菌分离鉴定及药敏试验,并结合病毒检测、病毒基因遗传进化及抗原表位分析,全面了解PRV毒株及与其混合感染的病原种类、致病性、耐药性、遗传进化及抗原表位位置等。【结果】从患病仔猪肝脏中分离获得两株优势生长的革兰氏阴性杆菌,一株为大肠杆菌(命名为JL-1/Guangxi/2014),另一株为志贺氏菌(命名为JL-2/Guangxi/2014),均为多重耐药菌株,仅对庆大霉素和阿奇霉素高度敏感。病毒检测结果显示PRV为阳性,命名为GXJL/China/2014(Gen Bank登录号KM386685);该PRV毒株与国内2012~2013年分离获得的PRV毒株聚为一支,而与国外Nia-1(FJ605136.1)等毒株亲缘关系较远;其抗原表位与PRV经典强毒株Min-A相比,在第48位插入一个Asp,且有5个氨基酸位点发生变异,造成部分抗原表位偏移。【结论】广西金陵某种猪场2014年发生的疫情是由PRV变异株与致病性大肠杆菌、志贺氏菌混合感染所致,可选用庆大霉素和阿奇霉素进行防治。

摘  要:
【Objective】The present study was conducted to diagnose causes of disease in a pig farm of Jinling, Guangxi in 2014 and investigate mixed infection trends of porcine pseudorabies virus(PRV) mutant and bacteria, in order to provide references for preventing effectively PR epidemic and formulating control measures. 【 Method 】 Bacteria of diseased was isolated and identified, and its drug sensitivity was detected with conventional laboratory detection methods.Then, the pathogenicity, drug resistance, genetic evolution and antigenic epitope location of PRV strain and mixed pathogens were fully investigated combined with virus detection, virus genetic evolution and antigenic epitope analysis.【Result】The results showed that two strains of Gram-negative bacterias with growth advantage were isolated from diseased piglet 's liver, one of them was Escherichia coli( named JL- 1 / Guangxi / 2014), another was Shigella( named JL- 2 /Guangxi/2014). Both of them were multi-drug resistant strains, and were only highly sensitive to gentamicin and azithromycin. In addition, the virus test results showed that PRV was identified to be positive, which named as GXJL/China/2014(Gen Bank accession No. KM386685). And this PRV mutant was clustered into a branch with PRV strains isolated from China from 2012 to 2013,while, had rather distant phylogenetic relationships with foreign Nia-1(FJ605136.1) and other strains. As compared with epitopes of classical PRV strain Min-A, a Asp was inserted into the 48 thlocus of epitopes of detected PRV mutant, and there were 5 mutated amino acids, resulting in partial epitope shift. 【Conclusion】The epidemic of a pig farm in Jinling, Guangxi in 2014 was caused by mixed infection of PRV mutant, pathogenic E. coli and Shigella, which can be prevented and treated with gentamicin and azithromyc.
