大花萱草“东方不败”的组织培养技术研究

摘  要:
以大花萱草"东方不败"的子房为外植体进行了组织培养的研究,结果表明,"东方不败"子房愈伤组织诱导分化的最佳培养基为MS+4 mg/L6-BA,不定芽分化率可达38.2%;在6-BA质量浓度为3 mg/L,IBA为0.5 mg/L时,繁殖系数可达6.4;适宜的生根培养基为1/2 MS+NAA 0.2 mg/L+IBA 0.2 mg/L+蔗糖(或白糖)20 g/L+琼脂粉6.5 g/L,每株试管苗生根数可达4~5条。组培过渡苗栽培基质为粗河沙,条件控制温度为24~28℃,相对湿度为85%~90%,并逐渐增加光照,移栽成活率可达92%。

摘  要:
In the article tissue culture was studied using ovary as explant and industrial seedling raising technique of "Dongfangbubai" H.hybrida.The results showed that the best culture medium for"Dongfangbubai" callus differentiation was MS,the concentration range of 6-BA 4 mg/L and adventitious bud differentiation rate reaches 38.2%.When 6-BA concentration was 3 mg/L and IBA 0.5 mg/L,propagation coefficient would be possible to reach 6.4.The suitable rooting medium was 1/2 MS+NAA 0.2 mg/L+IBA 0.2 mg/L+sucrose(white sugar) 20 g/L+agar 6.5 g/L,and the rooting number of each tube seeding reached 4~5 roots.The substrate for transitional cultivation of tissue culture seedling was thick riversand,the survival rate reached 98%,when the temperature was 24~28 ℃,the relative humidity was 85%~90% and the light intensity increased gradually,survival rate of tube seeding reached 92%.
