巨峰葡萄实生苗阶段发育过程中核酸含量的变化

摘  要:
实生个体的阶段发育包括不同发育时期和空间的生理生化变化。Kessler和Monse-llse(1961)研究了苹果、柑桔叶龄对其叶片中RNA含量及RNase活性的影响,均是成年树叶片的RNA水平高于幼年树,且随树龄而增加,与RNase活性随树龄而降低相一致。在豆梨、西洋梨、杜梨及英国常春藤上的有关研究亦有类似结果,成年组织与幼年组织的DNA水平差异不大,而RNA比值随实生个体的成熟而提高。这种有关代谢方面的研究在果树方面报道不多,在葡萄方面更少。本文对巨峰实生苗阶段发育过程中叶内遗传物质含量的变化进行了研究。 材 料 与 方 法 1.自根苗 巨峰葡萄于1986年春播种于温室,株行距1.5×2.0m,一般肥水管理,未进行修剪。1988年末首次开花,此时取样分析。取样方法:以卷须首次出现节位和开花的最低节位作为两个基本节位Ⅱ和Ⅳ,在Ⅱ与Ⅳ的中点取Ⅲ,Ⅱ以下10节为Ⅰ,Ⅳ以上10节和20节分别为Ⅴ和Ⅵ,在这6个部位分别取发育中庸的营养枝中部叶片,洗净,去除粗叶脉。单株重复3次。取样节位为所取叶片在营养枝上的节位加该营养枝在主蔓上的节位。 2.嫁接苗 2年生白香蕉扦插苗于1986年冬盆栽,一般管理,未修剪。1987年3月基部留17cm剪断,所发新蔓于6月中旬留4—6片叶剪断进行嫩枝嫁接。

摘  要:
The amounts of DNA, RNA and total nucleic acids, the RNA/DNA ratio in leaves of three-year-old 'Kyoho' self-rooted seedlings which had just entered into the reproductive state and grafted seedlings on Bei-xiangjiao rootstock which remained vegetative were determined in the course of their development.The effect of extended illumination ( 40w/m2 1830-0630 ) on the amount cf the above-mentioned contituents in the grapevine was investigated. It was found that starting from the basal end there was a progressive increase of leaf DNA along the stem up to the blossoming node, followed by levelling out of DNA.The peak values of RNA/DNA ratio, RNA total nucleic acid contents occurred in the leaf at the node where the first tendril appeared. The DNA content of the developmentally advanced portion was significantly increased by extended daily illumination.Trend of RNA/DNA ratio as well as RNA, total nucleic acid content in the leaves along the stem of the self-rooted seedlings below the lowest blossoming node was similar to that of the vegetative grafted seedlings.
