马铃薯对马铃薯块茎蛾取食危害的生理响应

摘  要:
【目的】明确云南马铃薯主栽品种合作88和青薯9号对马铃薯块茎蛾危害胁迫的生理响应,为抗虫马铃薯品种筛选及抗虫机制研究提供科学依据。【方法】室内盆栽合作88和青薯9号马铃薯,人工接种健康的马铃薯块茎蛾初孵幼虫,分别于取食危害胁迫0、3、6、12、24、48、72和96 h时进行马铃薯叶片取样,每处理重复3次;采用紫外分光光度计法测定不同取食胁迫时段马铃薯叶片丙二醛(MDA)、可溶性糖和过氧化氢(H_2O_2)含量及主要防御酶超氧化物歧化酶(SOD)和过氧化物酶(POD)活性。【结果】在马铃薯块茎蛾取食胁迫的3~96 h,两个马铃薯品种叶片的MDA和H_2O_2含量均增加,受害合作88和对照叶片的MDA平均含量分别为8.24±1.55和5.61±0.66 nmol/gFW,H_2O_2平均含量分别为2.92±0.16和2.14±0.11 U/gFW;受害青薯9号和对照叶片的MDA平均含量分别为9.60±0.58和4.62±0.55 nmol/gFW,H_2O_2平均含量分别为3.03±0.21和2.45±0.14 U/gFW;其中合作88叶片MDA和H_2O_2含量达最高值的时间(取食胁迫24 h)均早于青薯9号,青薯9号则分别在取食胁迫96和48 h时达最大值。虫害后,合作88叶片可溶性糖平均含量略低于对照,青薯9号叶片可溶性糖平均含量略高于对照。两个品种中POD活性均略低于对照,其中受害合作88和对照叶片POD平均活性分别为143.55±21.21和144.41±25.43 U/gFW,青薯9号和对照叶片POD平均活性分别为53.17±3.65和67.99±2.34 U/gFW。虫害后,合作88和青薯9号叶片SOD活性均高于对照,其平均活性分别增长了59.13%和20.61%。【结论】云南马铃薯主栽品种合作88对马铃薯块茎蛾取食危害胁迫的生理响应强于青薯9号。

摘  要:
【Objective】To verify the physiological responses of two major varieties of potatoes(Hezuo 88 and Qingshu No.9)planted in Yunnan after being damaged by potato tuber moth(PTM)and provide scientific basis for screening of insect-resistant variety and the mechanism of potato resistance to insects.【Method】Potted potato cultivars Hezuo 88 and Qingshu No.9 in Yunnan were inoculated with healthy potato tuber moth larvae for 0,3,6,12,24,48,72 and 96 h of feeding stress and then sampled,each treatment with three repetitions. The content of malondialdehyde(MDA),soluble sugar,hydrogen peroxide(H_2O_2)and the activities of superoxide dismutase(SOD),peroxidase(POD)in potato leaves during different feeding stress periods were measured by UV spectrophotometry.【Result】The content of MDA and H_2O_2 in the leaves of the two varieties increased within 3-96 h of feeding stress. The mean contents of MDA in the damaged Hezuo 88 and control were 8.24±1.55 and 5.61±0.66 nmol/gFW,respectively,and the mean contents of H_2O_2 were 2.92±0.16 and 2.14±0.11 U/gFW,respectively. The mean contents of MDA of damaged Qingshu No.9 and control were9.60±0.58 and 4.62±0.55 nmol/gFW,respectively,and the mean contents of H_2O_2 were 3.03±0.21 and 2.45±0.14 U/gFW,respectively. The occurrence of highest contents of MDA and H_2O_2 of Hezuo 88 were earlier than Qingshu No.9,being 24 and 24 h of feeding stress respectively,while Qingshu No.9 were 96 and 48 h of feeding stress,respectively. The average content of soluble sugar in the leaves of Hezuo 88 was slightly lower than that of the control,and the average content of soluble sugar in the leaves of Qingshu No.9 was slightly higher than that of the control,while the activity of POD in the two varieties was slightly lower than that of the control. The average activities of POD in leaves of damaged Hezuo 88 and the control were 143.55±21.21 and 144.41±25.43 U/gFW respectively. The average POD activities in leaves of damaged Qingshu No.9 and control were 53.17±3.65 and 67.99±2.34 U/gFW,respectively. The SOD activities in the leaves of Hezan 88 and Qingshu No.9 were higher than control,and the average activities of SOD in the two varieties increased by59.13% and 20.61% respectively.【Conclusion】The physiological response of main potato cultivar Hezuo 88 in Yunnan to the feeding damage of potato tuber moth is stronger than variety Qingshu No.9.
