摘 要:
将脱脂蚕蛹水解后用作产油微生物发酵培养基的氮源,生产营养价值高的可食用油脂,探究蚕蛹水解液中Fe~(3+)和Zn~(2+)对产油微生物解脂耶氏酵母(Yarrowia lipolytica)的生长及油脂累积的影响。首先于酵母对数生长期,在氮限制培养基中分别添加10-3mol/L Fe~(3+)和10-3mol/L Zn~(2+)溶液,使油脂合成量分别比对照提高了43.59%和38.70%,且同时添加Fe~(3+)和Zn~(2+)比单独添加Fe~(3+)和Zn~(2+)使油脂含量分别提高了27.88%、5.75%。然后于酵母对数生长期以脱脂蚕蛹水解液作为补料加入培养基,不仅使油脂的产量较酵母粉补料培养基提高了40.67%,而且使酵母合成油脂中不饱和脂肪酸与饱和脂肪酸(UFAs/SFAs)含量的比值从3.78提高到8.25,合成三酰甘油的sn-2和sn-1,3位脂肪酸的UFAs/SFAs值分别提高17.9%、179.8%。试验结果表明:以来源丰富的蚕蛹水解液作为解脂耶氏酵母发酵培养生产油脂过程中的补料,其浓度适中的Fe~(3+)、Zn~(2+)与其他有益组分对酵母生长和油脂合成及积累具有促进作用,并且使脂肪酸组成及分布改善。

The defatted silkworm( Bombyx mori) pupal hydrolysate was applied to cultivate oleaginous microorganisms as nitrogen source for producing high value-added edible oil. The effect of Fe~(3+)and Zn~(2+)in defatted silkworm pupal hydrolysate( DSWPH) on the growth and lipid accumulation of oleaginous microorganism Yarrowia lipolytica was explored.At exponential growth phase of yeast cells,10- 3mol / L Fe~(3+)and 10- 3mol / L Zn~(2+)were added to the nitrogen limited medium. The yield of lipid was increased by 43. 59% and 38. 70% compared with the control,respectively. The simultane-ous addition of Fe~(3+)and Zn~(2+)increased the lipid content by 27. 88% and 5. 75% than addition of Fe~(3+)and Zn~(2+)separately. Replacing yeast extract with DSWPH as added nutrient in medium for culture of yeast cells during the exponential phase,the yield of fat increased by 40. 67%,and the ratio of unsaturated fatty acids to saturated fatty acids( UFAs / SFAs) increased from 3. 78 to 8. 25. In addition,UFAs / SFAs value of fatty acids used to synthesize triglyceride on sn-2 position and sn-1,3 position increased by 17. 9% and 179. 8% respectively. The results indica-ted that using abundant DSWPH as added nutrient in culture medium of Y. lipolytica to produce fat,the moderate concentration of Fe~(3+),Zn~(2+)and other useful compositions in the hydrolysate could promote the growth,lipid synthesis and lipid accumulation of Y. lipolytica,and improve the composition and distribution of fatty acids.
