摘 要:
【目的】探研肉鸡IL8RB基因变异对肉质性状的遗传效应.【方法】采用PCR产物直接测序和结合序列比对软件筛查鉴定IL8RB基因外显子2遗传变异,分析其与‘黔画乌鸡’肉质性状的相关性.【结果】在‘黔画乌鸡’IL8RB基因外显子2中发现4个中度多态的同义突变位点g.22589317 C>T、g.22589423 C>T、g.22589485 C>T和g.22589476 T>C,4个SNP位点均产生3种基因型,基因型分布均未偏离Hardy-Weinberg平衡;g.22589317 C>T位点对嫩度的影响达到显著水平(P<0.05),基因型TT个体的嫩度值显著高于基因型CC个体(P<0.05);g.22589423 C>T和g.22589476 T>C位点均对粗脂肪含量的影响达到显著水平(P<0.05),g.22589423 C>T位点的基因型TT个体显著高于基因型CT个体(P<0.05),g.22589476 T>C位点的基因型CC个体显著高于基因型CT和TT个体(P<0.05).【结论】综合各基因型各指标之间的差异,揭示g.22589317C>T位点的CC基因型、g.22589423 C>T位点的TT基因型和g.22589476 T>C位点的CC基因型是改善肉质的有利基因型,可能作为提高肉鸡肌肉品质的分子遗传标记.

摘 要:
【Objective】 Interleukin-8 receptor bata(IL8 RB) plays a crucial role in the inflammatory response of livestock and poultry.The purpose of this study is to explore the genetic effect of IL8 RB gene variation on meat quality traits of broiler chickens.【Method】 PCR products direct sequencing and sequence alignment software were used to screen and identify the genetic variation of IL8 RB gene exon 2,and analyze its correlation with meat quality traits of Qianhua Black-bone chicken.【Result】 The four SNPs of g.22589317 C>T,g.22589423 C>T,g.22589485 C>T and g.22589476 T>C were detected in IL8 RB gene exon 2 in Qianhua Black-bone Chicken,and belonged to moderate polymorphism and synonymous mutation of each SNP.Three genotypes were produced,and its genotype distribution did not deviate from the Hardy-Weinberg equilibrium for each SNP.The g.22589317 C>T locus had significant effects on tenderness(P<0.05),and TT genotype was significantly higher in tenderness than CC genotype(P<0.05).The g.22589423 C>T and g.22589476 T>C loci had significant effects on crude fat content(P<0.05).The individuals of TT genotype at the g.22589423 C > T locus were significantly higher than CT genotype(P<0.05),and the individuals of CC genotype at the g.22589476 T>C locus were significantly higher than CT and TT genotypes(P<0.05).【Conclusion】 According to the differences among the indicators of each genotype of each SNP,all of those results suggest that the CC genotype of g.22589317 C>T,the TT genotype of g.22589423 C>T and the CC genotype of g.22589476 T>C are favorable for improving meat quality,and they may be a molecular genetic marker for improving meat quality of broiler chickens.
