摘 要:
从大豆玉米间作体系中分离大豆与玉米内生菌,通过无机磷培养基筛选得到具有溶磷能力菌株,且菌株均来源于玉米,利用钼蓝比色法测定玉米内生溶磷菌液中有效磷含量,判断内生菌溶磷能力,最终从35株分离自玉米根、茎、叶中具有溶磷能力的内生菌中筛选出1株溶磷效果最强菌株Y-18,有效溶磷量可达447.59 mg·L-1。根据菌株形态特性、生理生化特征及16S rDNA序列系统发育学研究,初步鉴定Y-18菌株为罗旺醋杆菌(Acetobacter lovaniensis)。通过控制变量法改变培养条件后发现,在其他变量不变条件下,最适碳源为葡萄糖,最适氮源为硫酸铵,最适宜培养温度为25℃,最佳磷酸盐含量为7 g·L-1,最佳pH为5,其中碳源、氮源、磷酸盐含量、pH对溶磷效果影响显著。

摘 要:
The endophytic bacteria strains of soybean and maize were isolated from the intercropping system of soybean and maize and the strains with the ability of dissolving phosphorus were screened through the inorganic phosphorus medium. Measured the effective phosphorus content of maize endogenous soluble phosphorus bacteria liquid by using Molybdenum blue colorimetry to determine the endogenous bacteria dissolving phosphorus ability. Finally, 35 endogenous soluble phosphorus bacteria strains were isolated from the roots, stems and leaves of maize, from which one of the strongest phosphorus dissolving effect strain Y-18 was screened out and its soluble phosphorus amount could reach 447.59 mg?L-1. According to the morphological characteristics, the physiological and biochemical characteristics and the phylogenetic systematic analysis of 16 S rDNA sequence of strains,Y-18 strain was preliminarily identified as acetic acid bacteria genera(Acetobacter lovaniensis). By controlling variable method to change the culture conditions, it was found that the optimal carbon source was glucose, the optimal nitrogen source was ammonium sulfate, the optimal culture temperature was 25 ℃, the optimal phosphate content was 7 g?L-1 and the optimal pH was 5,when other variables were constant. The carbon source, nitrogen source, phosphate content and pH had significant effects on the soluble phosphorus.
