突变体黑羽鹌鹑MHC classⅠ第4～8外显子多态性

摘  要：
以突变体黑羽鹌鹑为试验动物,用优化煮沸法提取鹌鹑基因组DNA。根据日本鹌鹑MHC classⅠ的mRNA设计1对引物,采用PCR技术扩增突变体黑羽鹌鹑MHC classⅠ基因片段,运用DNAStar6.0软件对该序列进行分析,显示该片段大小为991bp,G+C含量较高为62.46%,A+T为37.44%,并检测到5个突变位点。利用Blast程序与日本鹌鹑mRNA比对,该片段上含有第4～8外显子和第4～7内含子,其中第4外显子上含有4个T/C突变位点。结果表明,突变体黑羽鹌鹑MHC classⅠ第4外显子具有多态性,这些多态性与其免疫功能的关系如何,有待进一步研究。

摘  要：
The genome DNA of black feather quails was extracted by optimized co oking extraction method.One pair of primers were designed according to the Cotu rnix Japonica MHC classⅠmRNA sequence.The quail MHC classⅠgene was cloned by PCR and sequenced.The DNAStar 6.0 software was uesd to analyzed the sequence.The results showed the sequence included 991 bp and that the content of GC base s was higher(62.46%) than AT bases(37.44%).The sequence had exon4-exon8 and int ron4-intron7 by Blast with the Coturnix Japonica MHC class mRNA sequence.Fiv e mutations were found in this sequence,including T/C four mutations in exon4 a nd this indicated that the MHC classsⅠgene was high polymorphism.But this pol ymorphism was not analyzed with the immunity in quail and need to deeply research in the future.
