多房棘球蚴成体未分化细胞的鉴定及其分布特征

摘  要:
利用有丝分裂分子标志———组蛋白H3(Ser10)标记多房棘球蚴成体未分化细胞(neoblast)。同时,利用成体未分化细胞可能的分子标志———miR-71和AGO-2,通过原位杂交方法分析其表达特征。结果显示,多房棘球蚴组蛋白H3阳性细胞没有明显的组织分布特征,而且数量少;AGO-2阳性细胞在组织分布和数量上与组蛋白H3阳性细胞相似,而miR-71阳性细胞在原头蚴中分布广泛,并且在不同发育阶段的原头蚴中miR-71的表达水平存在明显差异。这些研究结果表明,miR-71分子在多房棘球蚴成体未分化细胞中呈非特异表达,不能作为鉴定它的分子标志。

摘  要:
Using mitotic molecular marker histone H3(Ser10)as a biomarker,mitotic cells in Echinococcus multilocularis metacestode,also known as neoblasts,were labeled.Meanwhile,expression characteristics of the possible molecular markers of neoblasts,miR-71and AGO-2,were analyzed through in situ hybridization.In result,the number of the histone H3positive cells was less than as expected in E.multilocularis metacestodes and there were no obvious tissue distribution characteristics.AGO-2positive cells had the similar features in tissue distribution and quantity,but miR-71positive cells were distributed widely in protoscoleces,and differently expressed among different developmental stages of protoscoleces.These results showed that miR-71were not specifically expressed in the neoblasts.Therefore miR-71is not acted as a molecular marker for identification of E.multilocularis metacestode neoblasts.
