甘薯β-胡萝卜素含量的QTL定位

摘  要:
构建甘薯分子连锁图谱,并对重要农艺性状进行QTL定位,进而克隆相关性状的主效基因,是甘薯育种研究的重要方向。在甘薯的农艺性状中,高胡萝卜素含量是重要育种目标之一。本研究利用低胡萝卜素含量甘薯品种漯徐薯8号(母本)和高胡萝卜素含量甘薯品种郑薯20(父本)杂交,以F1分离群体240个株系为材料,构建分子连锁图谱。父本57个连锁群,图谱的总长度4 968.62 cM,标记间平均距离10.35 cM;母本64个连锁群,图谱总长度为5 313.67 cM,标记间平均距离10.76 cM。以测定的β-胡萝卜素含量为指标,用MapQTL4.0软件检测到17个与甘薯β-胡萝卜素含量相关的QTLs,其中10个定位在父本图谱上,7个定位在母本图谱上,分别解释甘薯β-胡萝卜素含量表型变异的33.1%~62.1%。研究结果有助于甘薯胡萝卜素合成相关性状基因的克隆及分子标记辅助育种体系的建立。

摘  要:
Constructing molecular linkage maps, mapping of QTLs for the important agronomic traits, and cloning its major genes, are the research direction of molecular breeding in sweetpotato. The high content of carotenes is one of the important agronomic traits in sweetpotato breeding. In this study, 240 individuals of F1population, which derived from a cross between sweetpotato cv.‘Luoxushu 8'(female, a low carotene content cultivar), and cv.‘Zhengshu 20'(male, a high carotene content cultivar), were used to establish the molecular linkage maps. The female parent map contained 64 linkage groups, and covered 5 313.67 cM with an average marker interval of 10.76 cM. The male parent map contained 57 linkage groups, and covered 4 968.62 cM with an average marker interval of 10.35 cM. 17 QTLs associated with beta carotene content of sweetpotato were identified by the MapQTL4.0 software, and 10 of 17 QTLs belonged to‘Zhengshu 20'map and another 7 QTLs belong to ‘Luoxushu 8'map, and these QTLs showed 33.1%~62.1% of the phenotypic variance, respectively. The results will play an important role in cloning correlated trait gene and building the molecular assistant breeding system in sweetpotato.
