HPLC法测定犬血浆中伊曲康唑浓度

摘  要：
应用高效液相色谱法(HPLC)测定犬血浆中伊曲康唑的浓度。选用6条健康犬,以4 mg/kg体重背部皮下注射给药,采用HPLC测定犬血浆中伊曲康唑浓度(ITZ),应用3p97软件处理数据,计算药代动力学参数。结果表明血浆ITZ线性范围为10～1 000 ng/mL,伊曲康唑血浆添加浓度10、100、500ng/mL的平均回收率分别为86.9%、88.4%、89.2%,RSD分别为2.8%,1.2%,0.2%。说明HPLC法简便,准确可靠,适用于伊曲康唑在犬体药动学中血药浓度测定。

摘  要：
Pharmacokinetics of itraconazole injection was studied in healthy dogs.Subcutaneous injection was administrated to six dogs in a dose of 4mg/kg and the plasma concentration of itraconazole was detected by HPLC.The data was analyzed with 3p97 computer program.The results showed that the linearity was obtained over the range of 10~1000ng/mL and the recovery was 86.9%,88.4% or 89.2% when the addition concentration of itraconazole was 10 ng/mL/,100 ng/mL or 500 ng/mL respectively.The RSD was 2.8%,1.2% or 0.2% respectively.It was concluded that the method was simple,accurate and reliable,and could be used for the pharmacokinetic studies of itraconazole in dogs.
