摘 要:
2017年8月昆明寻甸某养殖户从山东郓城购进30头肉牛,在购进1周后陆续发生以呼吸道症状为主的疾病,部分牛只伴有腹泻。为了尽快确诊病因,本试验进行了牛常见传染病的病原学检测及序列分析,同时开展了牛支原体和细菌的分离鉴定。结果从牛肺脏组织和气管分泌物中检测出牛腺病毒7型及牛支原体核酸阳性,并分离鉴定出1株牛支原体,命名为XDJY-YN-2017株,还分离鉴定出2株细菌。测定出的牛腺病毒7型蛋白酶基因序列与NCBI上GenBank中的参考毒株(登录号:X53989)序列同源性达99%。基于16S rRNA序列构建的系统发育树显示,牛支原体XDJY-YN-2017株与以色列的72242株,英国的393B08、268B07株,中国的Ningxia-1、HB0801、CQ-W70、08M、Hubei-1、NM2012株同属于一个进化分支,古巴的422株和PG45株属于一个进化分支,Donetta株单独为一个进化分支。2株分离菌株经细菌生化鉴定为弗格森埃希菌(ID%=99.9%,T=0.8)和克吕沃尔菌属细菌(ID%=76.3%,T=0.61)。药敏试验结果显示,弗格森埃希菌对阿莫西林/克拉维酸、多黏菌素E、大观霉素、庆大霉素、安普霉素、呋喃妥因敏感,克吕沃尔菌属菌株对多黏菌素E、安普霉素、呋喃妥因、大观霉素敏感。试验结果表明外购肉牛由于长途运输、饲草饲料更换、饲养环境改变等应激因素,导致了牛腺病毒7型、牛支原体和细菌混合感染发病及死亡。

摘 要:
In August 2017,30 beef calves were purchased from Yuncheng,Shandong province,by a farmer in Xundian county,Kunming city. One week after arrival,respiratory diseases characterized by respiratory symptoms appeared one after another and began to die. To diagnose the possible causes of disease,nucleic acid detection and sequence analysis of common infectious diseases of beef cattle,as well as bacterial isolation and identification,were carried out. The results showed that nucleic acid tests for bovine adenovirus type 7 and Mycoplasma bovis were positive and one M. bovis strain( named XDJY-YN-2017) was isolated and identified from bovine lung tissues and trachea secretions. Two bacterial strains were also isolated and identified from bovine lung tissues. The sequence of protease gene of bovine adenovirus type 7 was 99% homologous to the reference strain( Accession: X53989) in NCBI Gen Bank. A phylogenetic tree based on 16 S r RNA sequences showed that XDJY-YN-2017 belonged to the same evolutionary clade as Israel 72242,UK 393 B08,268 B07,China Ningxia-1,HB0801,CQ-W70,08 M,Hubei-1 and NM2012 stains; formed another evolutionary clade with PG45 and 422 strains from Cuba; and was in a separate evolutionary clade from Donetta strain. The two bacterial strains were identified as Escherichia coli Ferguson strain( ID% = 99. 9%,T = 0. 8) and Kluyvera species( ID% = 76. 3%,T = 0. 61).Drug sensitivity test showed that E. coli Ferguson strain was sensitive to amoxicillin/clavulanic acid,polymyxin E,spectinomycin,gentamicin,ampulin,furantoin,while Kluyvera bacteria were sensitive to polymyxin E,ampomycin,furantoin,and spectinomycin.Thus,due to stress factors such as long distance transportation,forage feed replacement and environment change,the outsourced cattle developed mixed infections with bovine adenovirus 7,bovine mycoplasma and bacteria,leading to respiratory disease and death.
