摘 要:
使用小麦纯系尤比拉和黑麦自交系 L155杂交,F_1用0.1%的秋水仙精溶液处理,再用小麦亲本同交,得到 B_2F_1及其自交后代 B_2F_2群体。用单株植物C-带核型鉴定技术,从 B_2F_1和 B_2F_2群体筛选适合的不规则非整倍体植株,即含有一套完整的小麦染包体和一至数条黑麦染色体,或者黑麦的端着丝粒染色体或易位的染色体的植株。比较这些植株的染色体组成和特定基因的表达,可以把特定基因定位于特定的染色体和染色体臂上。使用这种方法,把黑麦的5个抗小麦白粉病基因定位于6RS 上;引起小麦籽粒皱缩的基因和叶耳色素基因定位于4RS 上;株高基因和叶宽抑制基因定位于2RL 上;而6RL,5RS 上含有叶宽促进基因。本研究证明,使用不规则非整倍体作基因的染色体定位是一个简单而有效的方法。

摘 要:
An attempt was made to localizc the genes in rye using irregular ancuploid plants, which possessed 21 pairs of wheat chromosomes and a varying numbcr of rye chromosomes.An wheat yariety Jubilar was crossed with a rye inbred line L155.The wheat-rye F_1 hybrids were doubled using 0.1% colchicine solution,and then were back-crossed to the wheat parent in the following two generations.From the B_2F_1 and B_2F_2 populations the irregular aneuploid plants can be obtained when these plants were cytologically analyzed by means of a modified C-banding technique on a plant-by-plant basis.The results that some charaeteristies could exhibit only in some plants,dcpcnding on the rye'Chromosomes or Chromosome arms remained in the plants,indicated that corre- sponding genes couldbe chromosomally localized.With this method the gene for resistance to powdery mildew was localized on the 6RS,the genes for seed shrivelling and for purple auricle on the 4RS,the genes for plant height and for leaf breadth suppression on the 2RL and the genes for leaf breadth promotion on the 5RS,and 6RL.The results indicated that the technique described in this paper appears to be a simple method for gene chromosomal locations.
