油菜种子内生菌的检测及杀菌消毒处理方法

摘 要:
采用组织分离法检测油菜种子内生菌的带菌率和栖息部位,并进行杀灭内生菌高效杀菌剂的筛选。结果表明:油菜各品种带菌率都较高,供试的8份材料中,带菌率最低的是品系“084”,其菌落出现频率也达70.8%,带菌率最高的为甘油5号,菌落出现频率高达100%。油菜种子的内生菌群主要为Alternariabrassicae(Berk)Sacc.、A.brassicola(Sehw)Witts、Seclerotiniasclerotiorum(Lid)Barg等和多种内生细菌。油菜内生真菌既可栖息在种皮,也可栖息在胚中,而内生细菌则只栖息于胚中。采用不同杀菌剂37℃下浸种7h,发现500倍50%地菌杀净可湿性粉剂对油菜种子内生菌有很强的杀菌作用。

摘 要:
It is a quick effective method to indentify disease resistanc(tolerance) of rapeseed to Sclerotinia sclerotiorum (Lid) de Barg by means of aseptic seedlings, breeding aseptic seedlings by means of rapeseed seeds,but the contamination often occurs to rapeseed a septic seedlings breeding. This research is the first to adopt tissue isolation to make a survey of endophytes-bearing rate and endophytes roosting locations of different rapeseed varieties. High efficient fungicides screening has been made. The results indicate that the bearing rate of each rapeseed variety is comparatively high. Among the eight varielties select for identiication, 084 is the variety with the lowest bearing rate which is 70.8% and Ganyou-5 is the highest with a bearing rate of 100%. Endophytes in rapeseeds are mainly Alternaria brassicae (Berk) Sacc?A.brassicola ( Sehw) Witts?Seclerotinia seclerorum (Lid) Barg etc. and a variety of endophytic bacteria. By means of tissue isolation,the germ-bearing rate of different rapeseed varieties is surveyed by choosing the entire seedcoat and embryo as a unit of isolation. The location of endophytes in rapeseeds is ascertation for the first time. Endophytic fungi are able to live in both seedcoat and embryo,while endophytic bacteria could only live in embryo. Amony three rapeseed varieties,Ganyou-5 is the variety with the highest germ-bearing rate of seedcoat, i.e. 40.2%,and zhong R-888 is the one with the lowest germ-bearing rate of seedcoat i.e.14.7%. In term of the germ-bearing rate of embryo,zhong R-888 ranks the highest i.e.39.7%,and Ganyou-5 i.e. 12.2%. Seeds of M004 and Ganyou-5 treated with Dijunshajing is respectively 0 and 1.1%,which is much lower than that of either CK or rapeseed with 0.1% mercuric choloride.
