冬夏季长白猪精液中细菌多样性与精子质量特性关联分析

摘  要:
为探究冬夏季长白猪精液中细菌多样性变化和精子质量特性之间的相关性,利用16S rRNA基因高通量测序技术得到冬夏季长白猪精液中微生物的特征序列(ASV),计算机辅助精子分析系统(CASA)分析冬夏季精子活力参数指标的变化。结果显示:长白猪精液中细菌多样性在冬夏季之间差异显著(P<0.05),门水平冬季优势菌群为厚壁菌门(Firmicutes),夏季为变形菌门(Proteobacteria),占比分别为51.3%和74.3%;属水平冬季占比最高的为乳杆菌属(Lactobacillus),夏季为假单胞菌属(Pseudomonas),占比分别为32.1%和52%;冬季样品中高丰度的乳杆菌属(Lactobacillus)与精子繁殖潜能呈显著正相关(P<0.05),而夏季样品中高丰度的假单胞菌属(Pseudomonas)与精子繁殖潜能呈显著负相关(P<0.05)。提示:季节变化影响公猪精液中细菌多样性,冬夏季猪精液中细菌组成不同,长白猪精液中的优势菌属与精子质量和繁殖潜能可能存在相关性。

摘  要:
To investigate the correlation between changes in bacterial diversity and sperm quality characteristics of Landrace boar semen in winter and summer, the 16 S rRNA gene high-throughput sequencing technology was used to obtain microbial amplicon sequence variants(ASV) from Landrace boar semen in the winter and summer seasons. The changes in sperm motility parameters in winter and summer were analyzed by the Computer-Assisted Sperm Analysis System(CASA). The results showed that the bacterial diversities in the semen of Landrace boar were significantly different between winter and summer(P<0.05). At the level of the phylum, the predominant phylum in the winter level was mainly Firmicutes(51.3%), and the predominant phylum in summer was mainly Proteobacteria(74.3%). At the genus level, the highest proportion in winter was Lactobacillus(32.1%), and in summer was Pseudomonas(52%). In the winter samples, Lactobacillus in high abundance was significantly positively associated with the reproductive potential of sperms(P<0.05), while in the summer samples, Pseudomonas in high abundance was negatively associated with the reproductive potential of sperm(P<0.05). In conclusion, seasonal variation affected the bacterial diversity in boar semen, and the bacterial composition in the semen was different in winter and summer; therefore, the dominant bacteria in Landrace boar semen might be correlated with their sperm quality and reproductive potential.
