摘  要:
对取自厦门海域养殖鱼排的真鲷以0.8、1.4、2.0μg.L-1不同浓度苯并[a]芘(BaP)分别进行4、24h的暴露染毒,用改进的慧星实验技术研究了BaP对真鲷血细胞DNA的损伤。结果表明:(1)通过慧星实验获得的DNA损伤指标显示,与空白对照组相比,暴露染毒组血细胞DNA均受到损伤且损伤存在显著性差异(P<0.01),剂量-效应关系明显,DNA损伤随着染毒时间的延长明显增加。(2)优化了适合检测真鲷血细胞DNA损伤的彗星实验电泳条件:20V、300mA、20min。(3)在样本数量较多的情况下,用细胞培养板板盖铺单层胶的彗星实验检测方式与用载玻片铺3层胶的“三明治”方式相比,具有操作简便、节省时间、制胶过程凝胶不易破损、重复性好等优点。

摘  要:
Live Pagrosomus major(Red seabream) obtained from a fish farm in Xiamen(Fujian,China) were exposed to different concentrations of benzo [a] pyrene(0.8 μg·L-1,1.4 μg·L-1 and 2.0 μg·L-1) for 4 h and 24 h respectively.DNA damage in blood cells of Pagrosomus major was evaluated by the modified comet assay.The results showed:(1) DNA damage evaluated by an index,was observed in blood cells of all exposed groups at B(a)P exposing time of 4 h and 24 h.There existed a significant difference in DNA damage between the exposed groups and the control(P<0.01).A dose-effect relationship of benzo [a] pyrene was observed on DNA damage levels in blood cells,which increased with the exposure time.(2) The optimum electrophoresis conditions for the comet assay were 20 V,300 mA for 20 min in order to better evaluate the DNA damage in blood cells.(3) A single low melting-point agrose on the cover of cell culture plates was developed and can be used for analysis of a large quantity of samples exposed to B(a)P.This method is more convenient,time-saving than the "sandwich" method for evaluating DNA damage on frosted microscope slides.
