HPLC-RID测定甘蔗茎节蔗糖、葡萄糖和果糖含量

摘  要:
研究高效液相色谱示差折光检测法(HPLC-RID)测定甘蔗茎节3种主要糖分蔗糖、葡萄糖和果糖的含量,摒弃有毒流动相乙腈,建立一种无毒、快捷、准确的检测方法.该法通过乙醇浸提,采用Waters Sugar PakⅠ色谱柱(6.5 mm×300 mm,10μm),流动相为超纯水,流速0.4 m L·min-1,柱温90℃,检测器温度35℃.检测方法线性关系好、灵敏度高,RSD为2.77%-4.5%(n=5),方法重复性良好;加标回收率高,达95.4%-101.8%;能满足甘蔗及相似作物中3种糖的准确分析.

摘  要:
Sucrose,glucose and fructose in sugarcane internodes were determined by high performance liquid chromatography with refractive index detector( HPLC-RID). The chromatographic separation was achieved on an Waters Sugar Pak I column(6. 5 mm ×300 mm,10 μm),which was held at a constant temperature of 90 ℃,using a mobile phase composed of pure water at a flow rate of 0. 4 m L·min- 1. RID temperature was set as 35 ℃. Average spike recoveries for 6 replicate determinations of three saccharides were found in range of 94. 2%- 101. 5%,with a relative standard deviation( RSD) between 2. 77% and 4. 5%.
