中华蜜蜂(Apis cerana cerana)抗中囊病选育研究

摘  要:
以江西省高安、上饶、靖安、遂川、南昌定地饲养的中蜂为素材,人工饲喂感染中蜂囊状幼虫病毒。从受试蜂群中选择抗病力强的蜂群,培育蜂王和雄蜂,作为中蜂子1代。然后用同样方法进行人工饲喂感染,培育中蜂子2代和子3代。再用中蜂与意蜂营养杂交方法来培育中蜂子4代、子5代和子6代,并测定了其感染发病率、产育力及7个工蜂形态指标;利用14条随机引物检测了其DNA多态性,并计算了遗传距离和进行聚类分析。结果表明:选育的中蜂子3代感染发病率显著低于普通中蜂,中蜂子6代感染发病率显著低于中蜂子3代;中蜂子6代蜂群的产育力比子3代和普通中蜂分别提高6.56%和10.22%,都达到显著差异;子4代、子5代、子6代工蜂的吻长、右前翅长度、右前翅宽度、第3腹节背板长度、初生重与亲本工蜂、子1代、子2代、子3代工蜂之间差异显著;所有子代工蜂的肘脉指数和右后翅的翅钩数与亲本工蜂之间差异不显著;RAPD标记显示中蜂子6代与亲本中蜂之间的遗传距离有较大差异。

摘  要:
Feeding the sacbrood virus to the sedentary colonies of Apis cerana cerana from different zones(Gaoan,Shangrao,Jing'an,Suichuan and Nanchang) of Jiangxi Province,and the colonies with strong resistance were chosen to breed the queen bee and the drone as the first generation of Apis cerana cerana(F1).In the same way,F2 and F3 were bred.Then through nutritional crossbreeding between Apis cerana cerana and Apis mellifera ligustica F4,F5 and F6 were bred.The incidence of the disease,the fecundity and seven morphological indexes were measured;also the polymorphism was tested by using 14 RAPD primers and the genetic distance was calculated and cluster analyzed.The result indicated:the infection rate of F3 was significantly lower than that of ordinary colonies of Apis cerana cerana,the infection rate of F6 was lower than that of F3,the fecundity of F6 was 6.56% higher than that of F3,and 10.22% higher than that of ordinary colonies of Apis cerana cerana.There were significant differences in the proboscis length,the width and length of right forewing,the length of 3nd tergum,the birth weight between of F4,F5,F6 and the parents,F1,F2 and F3;there was no significant difference between the parents and the offsprings in the brachium index and the number of wing claw.There was difference between F6 and the parents in the genetic distance of RAPD mark.
