番石榴叶水提物对拟穴青蟹抗病毒病能力及部分血清免疫因子的影响

摘  要:
【目的】研究番石榴叶水提物对拟穴青蟹抗病毒病能力及部分血清免疫因子活力的影响,为有效提高拟穴青蟹抗病毒病能力及促进拟穴青蟹养殖业健康发展提供理论依据。【方法】以番石榴叶水提物浸泡携带病毒的拟穴青蟹为试验组、不浸泡番石榴叶水提物为对照组,对比两组拟穴青蟹的成活率、病毒检出率及酸性磷酸酶(ACP)、碱性磷酸酶(AKP)、溶菌酶(LSZ)、超氧化物歧化酶(SOD)和酚氧化酶(PO)活性。【结果】经番石榴叶水提物浸泡处理的拟穴青蟹平均成活率为53.3%,较对照组提高50.1%,且死亡拟穴青蟹未检测出呼肠孤病毒和双顺反子病毒-1,而对照组死亡拟穴青蟹的这两种病毒检出率分别为100%和60%。随着试验时间的延长,拟穴青蟹血清ACP、AKP、SOD和LSZ等免疫相关酶活性均呈先升高后降低的变化趋势,其中试验组拟穴青蟹血清AKP和LSZ活性于用药后24 h时达最大值(7.5金氏单位/100 m L和5.1 U/m L),且均显著高于对照组(P<0.05,下同),ACP活性于用药后48 h时达最大值(2.8金氏单位/100 m L),SOD活性于用药后64 h时达最大值(27.1 U/m L),两者也均显著高于对照组;用药后72 h时,对照组拟穴青蟹PO活性急剧升高,显著高于试验组,但其余时间点与试验组间差异不显著(P>0.05)。【结论】番石榴叶水提物能增强拟穴青蟹的免疫功能,提高对青蟹病毒病的抵抗能力,且在浸泡后48 h内效果明显。

摘  要:
【Objective】In order to theoretical basis for improving antiviral ability of Scylla paramamosain and promoting healthy development of S. paramamosain aquaculture, the effect of aqueous extract from leaf of guava(Psidium guajava) on antiviral ability and some immunological factors activity in the serum of S. paramamosain. 【Method】S. paramamosain samples carrying virus were divided into two groups, one group was soaked in aqueous extract guava leaf as treatment group, another group wasn't be soaked as control group. Then the survival rate, detection rate of virus and activities of acid phosphatase(ACP), alkaline phosphatase(AKP), lysozyme(LSZ), superoxide dismutase(SOD) and polyphenol oxidase(PO) of two groups were compared. 【Result】The results showed that, the average survival rate of treatment group was53.3%, which was increased by 50.1%, compared with that of control group. The reovirus and bicistronic virus-1 were undetected from dead S. paramamosain of treatment group, while the detection rates of these two viruses from dead S.paramamosain of control group were 100% and 60%, respectively. As the extension of treatment time, the activities of immune-related enzymes ACP, AKP, LSZ and SOD in S. paramamosain serum increased firstly and then decreased. Furthermore, the activities of AKP and LSZ in S. paramamosain serum of treatment group reached up to maximums(7.5 King unit/100 m L and 5.1 U/m L) at the 24 thhour after administration, which were significantly higher than those of control group(P<0.05, the same below). And the activities of ACP and SOD in S. paramamosain serum of treatment group reached up to maximums(2.8 King unit/100 m L and 27.1 U/m L) at the 48 thand 64thhours, respectively, which were significantly higher than those of control group. The PO activity in S. paramamosain serum of control group sharply increased at the 72 thhour after administration, which was significantly higher than that of control group, and there was nonsignificant difference in PO activity between treatment and control groups at other sampling time(P>0.05). 【Conclusion】The aqueous extract of guava leaf can improve immunologic function of S. paramamosain and viral disease-resistant ability of S.paramamosain.
