犀牛钩虫ITS1-5.8S-ITS2基因序列的对比分析

摘  要:
为了鉴定云南省昆明动物园非洲白犀牛(Ceratotherium simum)所感染的寄生虫种属,采集犀牛粪便、镜检、孵化幼虫,提取虫体DNA,PCR扩增ITS1-5.8S-ITS2序列,测序后用SeqMan11.1软件拼接。在线Blast比对分析,序列上传NCBI,将其与已公布的钩虫ITS1-5.8S-ITS2基因序列用MEGA6.06软件构建系统进化树。结果显示,在犀牛粪便中发现虫卵;PCR扩增其ITS1-5.8S-ITS2基因序列,测序结果显示,长度均为781bp,Blast比对显示,与Uncinaria cf.(HE962182)、Uncinaria sp.(HQ262066)同源性分别为99%和98%。因此确认从犀牛分离的钩虫属于钩虫科弯口属钩虫(Uncinaria)。研究结果可为野生珍稀动物的寄生虫病防控提供参考,也为粪便中钩虫的进一步分类、鉴定和遗传变异研究奠定了基础。

摘  要:
The aim of the study was to amplify the ITS1-5.8 S-ITS2 sequence of the digestive tract nematode in African white rhinoceros Ceratotherium simum by PCR,then useing those gene sequences to construct the phylogenetic tree to further identify the parasitic relationship and molecular genetic information.Ceratotherium simumfeces was collected from a zoo in Yunnan province,the larvae were hatched and harvested,and DNA was extracted and amplified by PCR.The ITS1-5.8 S-ITS2 was sequenced and then assembled with SeqMan11.1 software.On-line Blast alignment of the ITS1-5.8 S-ITS2 gene sequence of hookworm was analyzed using MEGA6.06 software to construct the phylogenetic tree.The results showed that the length of ITS1-5.8 S-ITS2 gene was 781 bp.The homologies with:Uncinaria cf.(HE962182)and Uncinaria sp.(HQ262066)were 99% and 98%respectively.The results comfirmed,that isolated nematode from the feces of the African white rhinoceros(Ceratotherium simum)belongs to the genus Uncinaria.It provided reference for the control of parasites in wild rare animals and for the further classification,identification and genetic variation of hookworm.
