摘  要:
【目的】探究奶牛临床乳房炎中血红素氧合酶/一氧化碳(HO/CO)的作用及意义.【方法】采集泌乳期健康(对照组,Control, C)和患临床型乳房炎(试验组,Mastitis, M)荷斯坦奶牛乳腺组织,利用化学法测定内源性CO含量;H&E染色、实时荧光定量PCR(quantitative Real-Time PCR,qRT-PCR)、免疫蛋白印迹(Western blot)和免疫组织化学染色分析乳腺组织病理变化和HMOX1、HHMOX2的表达差异.【结果】采集的乳腺组织样品符合试验要求;两组样本中均有内源性CO、HMOX1、HMOX2基因和蛋白的差异性表达;HMOX1和HMOX2蛋白主要表达于乳腺上皮细胞;与C组相比,M组乳腺组织中内源性CO含量明显增加(P<0.01),HMOX1基因和蛋白表达显著上调(P<0.05)而HMOX2基因和蛋白表达显著下调(P<0.05).【结论】HO/CO系统参与奶牛临床型乳房炎的发生与发展,其可能调控乳腺上皮细胞的功能,发挥抗炎和细胞损伤保护等作用.

摘  要:
【Objective】 This study aimed to explore the role and significance of heme oxygenase/carbon monoxide(HO/CO) in clinical mastitis of dairy cows.【Method】 Breast tissues from healthy(control group, Control, C) and clinical-mastitis(experimental group, Mastitis, M) Holstein cows during lactation were collected, followed by determining the content of endogenous CO and analyzing the pathological change of breast tissue and expression differences of HMOX1 and HHMOX2 by the methods of H&E staining, quantitative Real-Time PCR,Western blot and immunohistochemical staining.【Result】 The sampled breast tissues met the requirements for the test.Both groups of samples had differential expression of endogenous CO,HMOX1,HMOX2 genes and proteins.HMOX1 and HMOX2 proteins were mainly expressed in breast epithelial cells.Compared with group C,the endogenous CO content in breast tissue of the M group was significantly increased(P<0.01),the expression of HMOX1 gene and protein were significantly up-regulated(P<0.05),and the expression of HMOX2 gene and protein were significantly down-regulated(P<0.05).【Conclusion】 The HO/CO system was involved in the occurrence and development of clinical mastitis in dairy cows.It might regulate the function of mammary epithelial cells, and played a role in anti-inflammatory and cell damage protection.
