蚕蛹羧甲基壳聚糖对小鼠成纤维细胞增殖与生长的影响

摘 要:
手术后组织粘连与组织损伤部位纤维的增生有密切关系。研究蚕蛹羧甲基壳聚糖对小鼠成纤维细胞L929增殖与生长的影响,探索其用作预防组织粘连生物医用材料的可行性。体外试验结果表明,培养液中添加0.1 mg/m L蚕蛹羧甲基壳聚糖能够极显著抑制小鼠成纤维细胞的增殖(P<0.01),抑制率达到50.08%±10.12%;荧光定量PCR检测小鼠成纤维细胞中的细胞生长因子TGF-β1和VEGF编码基因的表达极显著下调(P<0.01),并且这种抑制作用呈现明显的剂量效应。据此初步认为,蚕蛹羧甲基壳聚糖可有效控制成纤维细胞中关键的自分泌生长因子编码基因的表达,进而抑制机体的细胞增殖与生长以及炎症反应和血管增生,预防术后组织粘连。

摘 要:
Postoperative tissue adhesion is closely related to fibre hyperplasia in injured tissue. In this paper,the effect of silkworm( Bombyx mori) pupa carboxymethyl chitosan on proliferation and growth of L929 mouse fibroblast cells was detected,so as to explore feasibility of using it as biomedical material for preventing tissue adhesion. In vitro test showed that 0. 1 mg/m L silkworm pupa carboxymethyl chitosan could extremely significantly inhibit proliferation of mouse fibroblast cells( P<0. 01),with an inhibition rate of 50. 08%±10. 12%. qRT-PCR revealed that cell growth factor TGF-β1 and VEGF encoding genes in mouse fibroblasts were down-regulated at extremely significant level( P<0. 01). Besides,this inhibition showed an obvious dose-effect. It is preliminarily concluded that silkworm pupa carboxymethyl chitosan could effectively control gene expression of key autocrine growth factors in fibroblasts,thus further inhibit cell proliferation,inflammatory reaction and vascular proliferation,and prevent formation of tissue adhesion.
