三江黄牛mtDNA Cytb基因序列多态性及其系统进化分析

摘  要:
根据普通牛线粒体细胞色素b(Cytb)基因序列设计引物,扩增获得三江黄牛线粒体细胞色素b基因全序列,并以绵羊为外属对牛亚科代表性的17个品种的Cytb基因序列进行系统进化分析,以期了解三江黄牛的遗传多样性。结果表明:三江黄牛线粒体Cytb基因序列长1 140 bp,T、C、A、G 4种碱基的平均百分比分别为25.78%、28.55%、32.15%和13.52%;三江黄牛共有16种单倍型,平均单倍型多样性(Hd)为0.6923,平均核苷酸多样性(Pi)为0.006 25,遗传多样性较为丰富;系统进化分析显示三江黄牛是中国众多黄牛类群中的一支,与北方黄牛的亲缘关系较近,推测其是由普通牛和瘤牛共同进化而来,目前应该扩大种群的数量,以维持群体内部的遗传多样性。

摘  要:
The Sanjiang cattle mitochondrial Cytochrome b gene was cloned using a specific primers designed according to the mitochondrial DNA sequence of bovine, which was then used for phylogenetic analysis combined with the mt DNA Cytochrome b gene of 17 bovine typical cattle and cattle breeds of China, with that of sheep as the exogenous reference. These data may be beneficial for our understanding the genetic diversity of Sanjiang cattle. The results showed that, Sanjiang cattle mitochondrial Cytochrome b gene sequence length 1 140 bp. T, C, A, G took the average content of 25.78%, 28.55%, 32.15% and 13.52%, respectively; a total of 16 Sanjiang cattle haploid type with the average haploid type diversity(Hd) of 0.692 3, the average nucleotide diversity( Pi) of 0.006 25 were identified and Sanjiagn cattle have a abundant genetic diversity; Phylogenetic evolution analysis found that Sanjiang cattle is an independent cattle breeds, obtained the closest genetic relationship with Northern cattle, and presume to be co-evolved by the Bos Taurus and Bos indicus. Now the size of breeding population in Sanjiang cattle should be enlarged to maintain the genetic diversity level of population.
