牛产气荚膜梭菌分离株毒素基因的检测与分析

摘  要:
为了解黑龙江省牛源产气荚膜梭菌分离株的基因型,检测β2和CPE两种次要毒素因子在牛源产气荚膜梭菌分离株中的携带情况。通过PCR方法检测了产气荚膜梭菌13个野毒株的cpa、cpb、etx、itx四种分型毒素、cpb2、cpb2的两个等位基因突变体(atypical cpb2和consensus cpb2)及肠毒素(cpe)基因。结果表明:92.3%(12/13)的产气荚膜梭菌分离株为A型,检测到1株D型;A型菌中β2毒素基因检出率为66.7%(8/12);β2毒素基因阳性菌株中75%(6/8)携带非典型cpb2基因,50%(4/8)携带典型cpb2基因,非典型突变体占优势。动物致病性试验发现β2毒素阳性菌株(8株)与β2毒素基因阴性菌株(4株)各有2株分离株具有致病性。所有菌株CPE毒素基因PCR检测均为阴性。研究为产气荚膜梭菌病的发病机理和疫苗开发提供了理论依据。

摘  要:
In order to understand the genotypes of main toxin secreted by Clostridium perfringens isolates from cattle in Heilongjiang province, the correlation between the secondary toxins of β2 and CPE and the pathogenicity of Clostridium perfringens was determined. Four isolates of cpa, cpb, iap, and etx, and two alleles of cpb2 and cpb2(atypical cpb2 and consensus cpb2) and enterotoxin(cpe) gene were detected in 13 isolates of Clostridium perfringens by PCR. Results showed that 92.3%(12/13) of isolates was A-type, only one was D-type. 66.7%(8/12)of β2 toxin genes was detected in A-type; 75%(6/8)positive strains ofβ2 toxin gene carried the atypical cpb2 gene and 50%(4/8) carried the typical cpb2 gene, which meant atypical mutants were predominant. Animal pathogenicity tests showed that both positive and negative strains of β2 toxin genes had 2 isolates with pathogenicity. PCR test of all strains of CPE toxin gene were negative. A theoretical basis was provided for the pathogenesis of the disease and vaccine development.
