摘  要:
胡杨具有典型的异形叶性,是研究植物叶形发育的理想材料。但至今尚不清楚胡杨异形叶发生的分子调控机制。本研究以胡杨披针形和宽卵圆形幼叶为材料,利用qRT-PCR技术分析了参与叶基-顶轴发育的rot3和rot4基因、参与叶中-侧轴发育的an和spk1基因在胡杨异形叶发生中的表达模式,进一步分析了这些基因对胡杨异形叶发生的调控作用。发现rot3和spk1在披针形叶中的表达显著高于在宽卵圆形叶中表达,特别在叶形发育早期。而rot4和an则在宽卵圆形叶中的表达显著高于在披针形叶中表达,尤其是叶形发育早期。这暗示在胡杨叶形发育过程中,尤其是早期,rot3和spk1能促进胡杨叶形向披针形发育,而rot4和an则能促进胡杨叶形向宽卵圆形发育。同时也证明这4个预测基因在胡杨中是存在的,它们通过控制基-顶轴和中-侧轴极性建成调控胡杨异形叶的发生。

摘  要:
The poplar species Populus euphratica Oliv. has the typical heterophylly, is an ideal model species for studying leaf shape development of plants. However, it is unclear about the generative molecule mechanism of P.euphratica heterophylly. In this study, two typical leaves of P. euphratica, the lanceolate and the broadly ovate tender leaves within buds, were selected as research objects. Their shape changes were investigated at tissue level using unfolded piece coupled with freehand section method. Moreover, with quantitative real-time PCR(q RT-PCR) technology, the expression patterns of rot3, rot4, an, and spk1 in the above two kinds of leaves were analyzed, and further revealed the underlying generative mechanism of heteromorphic leaves of P. euphratica.Through comparing the differential expression of the above four genes in the lanceolate and the broadly ovate tender leaves, the expression of rot3 and spk1 in the lanceolate leaves was dramaticlly higher than those in the broadly ovate, but on the contrary, the expression of rot4 and an in the later was significantly higher than those in the former, what indicated that rot3 and spk1 can promote the leaf shape development of P. euphratica to lanceolate leaves, especially in the early period; however, rot4 and an can accelerate the leaf shape development of P. euphratica to oval leaves, especially in the early period too. It was suggested that rot3, rot4, an, and spk1 do exist in P. euphratica, and the genesis of heteromorphic leaves of P. euphratica is co-regulated by these genes controlling the development of apical-basal axis medial-lateral axis, particular ly in the early period of leaves germination within buds.
