锡金海棠Giemsa C带诱导及带型分析

摘 要:
对锡金海棠 ( Malussikkimensis)的 Giemsa C带的诱导技术改进及带型分析表明 ,其带型公式为 2 n=3 4 =3 CIT++ 1 CI+ T++ 1 CIT+ 2 CI+T++ 1 IT+3 CT++ 4T++ 6CI+ 7C+ 4I+ 2 ,主要特征以着丝粒带 ( C)为主 ,依次是中间带 ( I)和端带 ( T)第 9号染色体无带 ,2 ,3和 1 7号染色体带纹呈杂合态 ,Giemsa C带的异染色质含量为 3 2 .62 %。本文还介绍了获得木本植物小染色体 Giemsa C带的技术关键是带纹处理前的高温 ( 90℃~ 1 0 0℃ )处理。此外还探讨了其可能的机制

摘 要:
An improved method for Giemsa C band induction and the C banding patterns of Malus sikkimensis are reported in this paper. The C banding formula of this species is 2n=34=3CIT ++1CI+T ++1CIT+2CI +T ++1IT+3CT ++4T ++6CI+7C+4I+2, with centromeric bands being predominant, followed in sequence by intercalary and telomeric bands. There seem to be no bands in pair 9, while heterozygous bands are present in pairs 2, 3 and 17. The amount of heterochromatin is 32.62% in the Giemsa bands.The most important improvement in the method described here lies in a high temperature treatment (90℃~100℃) prior to BSG technique. It proves very useful for the production of chromosome C bands in woody plants. The possible underlying mechanism(s) are discussed.
