种子老化对扁蓿豆种质遗传完整性变化的影响

摘  要:
采用SSR分子标记对经老化处理的不同发芽率水平扁蓿豆种质进行遗传完整性分析,结果表明:(1)用10个有效引物对4个不同发芽率种质进行PCR扩增,共检测到25个等位基因,每个位点的等位基因数在1~4之间,平均为2.5个;(2)老化处理群体的多态性位点百分率、等位基因数、有效等位基因数、基因多样性指数、Shannon指数等遗传参数与对照相比都有所下降,表明老化处理后群体内的遗传多样性低于对照;发芽率为20%时,等位基因数呈显著性差异,说明扁蓿豆种质更新发芽率临界值在20%~40%之间;(3)SSR分子标记可以作为老化扁蓿豆种子遗传完整性变化的检测方法,对于异质种质资源材料,低的发芽率更新标准不利于种质资源遗传完整性的保持。

摘  要:
The effects of artificial aging on genetic integrity of Melilotoides ruthenica germplasm populations in different germination level were studied based on SSR markers.The results showed that 25 alleles amplified by PCR were detected with ten primers,number of alleles per locus was 1~4,mean number of alleles was 2.5;the percentage of polymorphic bands,the number of alleles(Na),effective number of alleles(Ne),Nei's gene diversity index(H) and Shannon's information index(I) were lower in artificially aged population than in high germination(CK) population based on SSR;the number of alleles was lower than that of CK population when germination rate of the population was 20%,and the difference was significant(P<0.05),which indicated that regeneration germination was 20%~40%;SSR molecular markers can be used as a method with which genetic integrity of Melilotoides ruthenica could be detected;a low germination percentage standard for regeneration is disadvantageous for maintaining the genetic integrity of germplasm resource.
