应用AllGlo探针检测对虾白斑综合征病毒研究

摘 要:
用AllGlo探针检测对虾白斑综合征病毒,阳性反应呈典型的S曲线,说明本检测探针和引物设计合理。经灵敏度测定,当样品DNA浓度低至100拷贝/μL时,仍可以有效扩增,但在103/μL之上时结果最佳。同时,经过与传统探针检测对比试验得出:用Taqman标记的探针检测相同量的样品时,AllGlo MAR标记的探针(FAM通道)CT值与FAM标记的探针的CT值没有明显区别,但是MAR标记的探针的荧光信号强度要比FAM标记的探针约高40%,更适合于对虾白斑综合征病毒的检测。

摘 要:
The latest fluorescent PCR probe—AllGlo probe was used to detect white spot syndrome virus(WSSV) in the study.The typical S-curve of positive reaction indicated that the probe and primers were designed correctly.The probe still worked effectively when the sample DNA concentration was as low as 102 copies/μL and the best result was got when the DNA concentration was above 103 copies/μL.The comparison with FAM probe indicated that the CT value of AllGlo MAR labeled probe(FAM channel) and FAM labeled probes were not apparently different when the sample DNA concentration was the same,but the fluorescence signal intensity of the AllGlo MAR labeled probe was about 40% which was higher than that of the FAM labeled probe,therefore the AllGlo probe was more suitable for the detection of WSSV.
