冠菌素对桑悬浮细胞次生代谢物产量的影响

摘  要:
桑树次生代谢产生的萜类、酚类和生物碱等具有多种生理活性,研究利用桑树细胞体外高效生产次生代谢产物的方法与技术,有助于实现对桑树药用活性成分的规模化开发利用。在建立桑悬浮细胞培养体系的基础上,向桑悬浮细胞系的培养基中添加不同浓度的植物生长调节物质冠菌素(COR),以促进细胞中次生代谢物的积累。试验结果表明,用5.0μmol/L COR诱导处理后桑悬浮细胞培养体系中总黄酮的质量浓度达到最高值(0.630±0.003)mg/m L,比对照组增加86.43%(P<0.05);1.0μmol/L COR诱导处理组桑悬浮细胞培养体系的1-脱氧野尻霉素(DNJ)、总酚质量浓度分别达到(23.50±0.49)mg/L、(56.71±1.52)mg/L,与对照组相比分别提高264%和50.27%(P<0.05);50.0μmol/L COR诱导处理组桑悬浮细胞培养体系的超氧化物歧化酶(SOD)活力达到4 595.3 U/g,显著高于对照组(P<0.05);几种浓度COR诱导对桑悬浮细胞培养体系的多糖积累均有显著抑制作用(P<0.05)。试验结果提示:COR作为诱导子能够增强桑悬浮细胞中总黄酮、总酚和生物碱类次生代谢物的合成与积累,且细胞的SOD活性亦显著提高,尤以低浓度(1.0μmol/L)COR的诱导效果最佳。

摘  要:
Terpenoids,phenols and alkaloids produced by mulberry secondary metabolism have various physiological activities. To meet the demand of large-scale development and utilization of medicinal active ingredients from mulberry,we should establish methods and techniques for efficient in vitro production of secondary metabolites from mulberry cells.On basis of establishing mulberry suspension cell lines,different concentrations of coronatine( COR) were added into mulberry cell suspension cuture to stimulate accumulation of secondary metabolites in the cells. The results showed that the concentration of total flavonoids in mulberry suspension cells reached the highest value [( 0. 630 ±0. 003) mg/m L]after induced by 5. 0 μmol/L COR,which was 86. 43% higher than that of the control group( P<0. 05).The content of 1-deoxynojirimycin( DNJ) and total phenol in experimental group supplemented with 1. 0 μmol/L COR reached( 23. 50 ±0. 49) mg/L and( 56. 71 ± 1. 52) mg/L respectively,being significantly increased by 264% and 50. 27% over the control group( P< 0. 05). When the concentration of COR was 50. 0 μmol/L,superoxide dismutase( SOD)activity of the treated group reached 4 595. 3 U/g in mulberry suspension cells, which was significantly higher than that of the control group( P<0. 05). After induced by various concentrations of COR,the accumulation of polysaccharides was significantly inhibited( P<0. 05). In conclusion,COR as an inducer could enhance the accumulation of secondary metabolites such as total flavonoids,total phenols and alkaloids,and increase activity of SOD in mulberry suspension cells. The best induction effect was achieved by using low concentration( 1. 0 μmol/L) of COR.
