REV感染对鸡NLRC5及其信号通路相关基因转录的影响

摘  要:
为探讨鸡NLRC5信号通路相关基因在禽网状内皮组织增生病病毒(REV)刺激后转录水平的变化,通过REV刺激鸡胚成纤维细胞(CEF),在刺激后7天采集细胞,提取总RNA,并设计相关引物,应用荧光定量PCR方法检测NLRC5信号通路相关基因(NLRC5、MHC-I、IL-6、IL-18、STAT1、STAT3、NF-κB)m RNA表达变化规律。结果表明:REV感染后MHC-I、IL-18、STAT1和STAT3的转录水平高于未刺激组,差异显著(P<0.05),NLRC5、IL-6和NF-κB转录水平低于未刺激组,差异不显著(P>0.05),研究初步证实REV刺激能够引起CEF产生免疫应答反应,且对NLRC5基因及其信号通路产生影响,REV可能通过抑制NLRC5基因表达来抑制机体先天性免疫。

摘  要:
This study was conducted to investigate the transcription change of related genes in chicken NLRC5 signal pathway stimulated by reticuloendotheliosis virus(REV),aiming to illustrate expression pattern of NLRC5-related genes and mechanism and function of chicken NLRC5 in innate and acquired immune,which lay a foundation for preliminary established NLRC5 regulation relationship pattern. REV was used to stimulate chicken embryo fibroblast(CEF)for168 hours. Total RNA was isolated from cells gathered at 0 and 168 h time points,then the transcription difference of association genes in NLRC5 signal pathway(NLRC5,NF-κB,MHC-I,IL-6,IL-18,STAT1,STAT3,NF-κB)was determined by quantitative real-time PCR. The results indicated that compared with the control groups,the expression levels of MHC-I,IL-18,STAT1 and STAT3 increased significantly in the infected groups at 7 days post-infection(DPI)(P<0.05).The expression levels of NLRC5,IL-6 and NF-κB decreased at 7 DPI(P>0.05). These results suggested that NLRC5 and some genes involved in the NLRC5 pathway could play a key role in antiviral immunity,and REV infection arouse innate immune suppression caused by inhibition of NLRC5 expression.
