熊猫粪便中纤维素降解菌的筛选与鉴定

摘  要:
为了探究熊猫消化道内高效的纤维素降解菌,实现高纤维类饲料资源化利用,本试验以熊猫粪便为研究对象,通过形态学观察,生理生化实验及16SrDNA鉴定,该菌株为革兰氏阴性好氧假单胞菌,并将其命名为NC020209。采用3,5-二硝基水杨酸显色法对菌株产酶条件进行研究,结果表明:在培养基的pH为6.0,温度为25℃,装液量30%,接种量4%,摇床转速为150rpm的条件下培养36h,该菌株的羧甲基纤维素酶活性达到最大24.362U。

摘  要:
To promote high fiber feed utilization and explore the cellulose degradation in panda's digestive tract,the strain of gram-negative aerobic pseudomonas belonging to the cellulose degrading bacteria was isolated from panda excrement,the cell morphology,physiological and biochemical experiments and16 srDNA appraisafurther colonized,observed and named as Pseudomonas poae RE*1-1-14,NC020209.Ferment condition of the extracellular cellulase were analyzed with 3,5-dinitrosalicylic acid and the results showed that enzyme activity of the strain peaked at 24.362 Uwhen the initial pH was 6,the temperature was 25℃,broth content was 40%,inoculum was 4%,rotation speed was 150r·min-1 after culturing 36 h.
