口蹄疫3A蛋白基因的表达及其抗原性研究

摘  要:
从pUCm-3ABC质粒切割FMDV的3A蛋白基因DNA,将其与pET-30c(+)载体连接,构建pET30C3A原核重组表达质粒,序列分析表明,pET30C3A重组质粒的插入3A蛋白基因的阅读框架正确。pET30C3A-BL21经IPTG诱导后可表达3A融合蛋白。Western Blot试验证明该表达蛋白具有较好的抗原活性,提示可进一步用重组表达的FMDV 3A蛋白作抗原检测PMD。

摘  要:
The DNA coding for 3A protein of foot and mouth disease virus was cloned into pET- 30c(+) expression vector, and the reconstructed plasmid pET30C3A was obtained, which could express the 3A protein with high efficiency . Western Blot test showed that the expressed 3A protein could react specifically with FMDV positive serum,indicating that pET30C3A plasmid could be used for FMDV diagnosis.
