山羊无浆体MSP4全长基因的克隆测序与B细胞抗原表位预测

摘  要:
该试验对重庆忠县感染山羊的无浆体主要表面蛋白4(MSP4)基因进行克隆测序,并对其系统进化、基因型以及推导蛋白的B细胞抗原表位进行分析.结果表明:所克隆的MSP4基因片段大小为870bp,CDS序列全长852bp,编码283个氨基酸,GenBank登录号为HQ840746.该序列与已公布的羊无浆体MSP4(AY702923,HQ456347,HQ661165)同源性最高,均达99.8%.系统发育分析发现,该序列被聚类到羊无浆体群.抗原表位预测表明忠县山羊无浆体MSP4蛋白的优势B细胞抗原表位在N端第77-82(AGTALS),93-98(APSLSG),216-220(TAGLV)和244-249(GSTLAI)4个区段.本研究从分子水平证实重庆存在羊无浆体感染,我国羊无浆体至少有5个基因型,重庆山羊无浆体MSP4蛋白具有4个优势B细胞抗原表位,为羊无浆体病亚单位疫苗的研发提供了参考.

摘  要:
In the study,we cloned and sequenced the major surface protein 4(MSP4)gene of Anaplasma ovis in goat from Zhongxian county,Chongqing,and then analyzed the B cell eptiopes on MSP4 of A.ovis.The results showed that the MSP4 sequence length of A.ovis was 870 bp and the complete CDS was 852 bp,which encoded 283 amino acids.Its GenBank accession number was HQ840746.The determined sequence was highly similar to that of AY702923,HQ456347 and HQ661165,with a homology of 99.8%.Phylogenetic analysis placed the sequence in the A.ovis cluster.B cell epitope prediction showed that there were four B cell eptiopes,77-82,93-98,216-220 and 244-249,on MSP4 of A.ovis.These results confirmed that infection of A.ovis occurred in goats in Chongqing.Analysis of all MSP4 sequences from A.ovis demonstrated that there were at least 5 A.ovis genotypes in China.
