摘  要:
将新城疫病毒(Newcastle disease virus, NDV)刺激鸡成纤维细胞(chicken embryo fibroblast, CEF)产生的外泌体(NDV Ex)静脉注射7日龄SPF雏鸡,通过体内成像观察NDV Ex在体内蓄积情况与主要分布器官,qPCR检测NDV Ex作用下肺脏、脾脏、肝脏病毒载量变化与Ⅰ型干扰素的表达量变化,分离肺泡巨噬细胞并进行形态学观察及表型鉴定,通过流式细胞术检测肺泡巨噬细胞在NDV Ex作用下吞噬能力的变化,评估NDV Ex在鸡体内对NDV增殖的影响。结果表明,雏鸡静脉注入NDV Ex后荧光主要分布于肺脏;qPCR结果显示NDV Ex可以促进NDV的体内感染,并抑制肺脏、脾脏与肝脏中Ⅰ型干扰素和肺泡巨噬细胞吞噬有关细胞因子(IL-1β、IL-6和iNOS)的表达;流式细胞术结果显示NDV Ex抑制了肺泡巨噬细胞的吞噬能力。

摘  要:
The Newcastle disease virus(NDV) stimulated exosome(NDV Ex) produced by the chicken embryo fibroblast(CEF) was injected intravenously into the 7-day-old SPF chick, in vivo imaging was used to observe the accumulation of NDV Ex in vivo and the main distribution organs.qPCR was used to detect the changes of viral load and the expression of type Ⅰ interferon in the lungs, spleen and liver under the effect of NDV Ex.Alveolar macrophages were isolated and identified by morphological observation and phenotype.The phagocytic ability of alveolar macrophages under NDV Ex was detected by flow cytometry to evaluate the effect of NDV Ex on NDV proliferation in chickens.The results showed that intravenous injection of NDV Ex fluorescence mainly distributed in the lung, qPCR results showed that NDV Ex can promote the infection of NDV in vivo and inhibit the expression of type Ⅰ interferon and alveolar macrophages phagocytosis related cytokines(IL-1β,IL-6 and iNOS) in the lung, spleen and liver.Flow cytometry showed that NDV Ex inhibited the phagocytosis of alveolar macrophages.
