土壤微生物在不同纤维素富集条件下的多样性

摘  要:
应用PCR-DGGE分子技术并结合聚类分析、主成分分析(PCA)等统计分析方法,对土壤微生物在几种不同的纤维素富集培养条件下的多样性进行分析。结果发现,不同的纤维素富集培养条件对土壤微生物的多样性有不同程度的影响:CMC和PCS两种培养基在50℃时所回收的土壤微生物的菌群结构和组成比较相似;纤维素富集培养基(J培养基)同CMC两种培养基所回收的土壤微生物菌群结构和组成却有很大的差异;50℃条件下所回收的土壤微生物其菌群结构和组成同28℃、37℃两个温度相比也相差较大。这一结果表明,组合不同的纤维素富集培养条件,结合分子和统计分析,可以对土壤样品在不同纤维素富集培养条件下微生物的多样性进行检测和评估,同时还可以为分离目标菌时富集培养条件的选择提供很有价值的参考。

摘  要:
The diversity of soil microbes cultured under different cellulose enrichment conditions was analyzed by PCR-DGGE fingerprinting combined with cluster analysis and PCA. Two soil samples were collected to make serial dilutions, inoculated into three kinds of cellulolytic microorganism enrichment media and incubated under three temperatures (28 ℃, 37 ℃, 50 ℃). The DNA of these cultured samples was extracted to amplify 16S rDNA gene V3 region, the products of PCR were used for DGGE analysis. According to the 16S rDNA V3-DGGE banding patterns, cluster analysis and PCA were performed by using software of GelCompar II and Matlab respectively. Both cluster analysis and PCA demonstrated that cellulose enrichment culturing conditions had different effects on the soil microbes. The structure and components of the samples incubated in CMC medium and PCS medium at 50 ℃ showed a relatively high level of similarity, in contrast, those incubated in CMC medium and J medium were significantly different from each other. And also there was a significant discrepancy of the diversity between the samples incubated at 50 ℃ and the samples incubated at 28 ℃ or 37 ℃. Different cellulose enrichment conditions combined with molecular techniques and statistical analysis may facilitate the detection and evaluation of microbial diversity in soil samples for cellulose degrading populations, and provide references for optimizing and selecting cellulose enrichment conditions.
