To examine the effect of Vascular Endothelial Growth Factor (VEGF) on the cleavage rate and development rate to 4 to 8-cell stage of Japanese Black oocyte, human recombinant VEGF 165 was supplemented at the different concentration of 0, 0.1, 1 and 10ng/ml during in vitro maturation (IVM), in vitro fertilization (IVF) and in vitro culture (IVC) in the present experiement. Cumulus oocyte complexes (COCs) were matured for 22 h in modified Synthetic Oviduct Fluid (m-SOF) supplemented with 10% fetal bovine serum (FBS), inseminated for 6 h in BO medium and cultured in 1% FBS-m-SOF. The cleavage rate was improved from 47.3% of the control to 51.4%, 72.6%(P<0.05) and 68.5%(P<0.05) in 0.1, 1 and 10 ng/ml VEGF groups respectively. The development rate to 4 to 8-cell stage was increased to 36.4%, 59.7%(P<0.05) and 52.9% from 32.0%(control) in 0.1, 1, 10 and 0ng/ml(control) groups respectively. These results indicate that VEGF has a beneficial effect on the development of Japanese Black oocyte.
