神蜂精对佐剂性关节炎大鼠的治疗作用

摘  要:
研究神蜂精对佐剂性关节炎大鼠的治疗作用,并探讨其作用机制。方法:60只SD雄性大鼠进行随机性分组,分为正常组、模型组、神蜂精高剂量组(1.0 mL/kg/d)、神蜂精中剂量组(0.5 mL/kg/d)、神蜂精低剂量组(0.25 mL/kg/d)、阳性组(复方醋酸地塞米松乳膏给药组,1.0 g/kg/d),每组10只。除正常组外,其余组大鼠右后足跖皮下注射完全弗氏佐剂诱导产生佐剂性关节炎。造模14 d后连续给药28 d。测量注射足足爪肿胀程度和免疫器官重量,酶联免疫吸附法(ELISA法)检测血清肿瘤坏死因子-α(TNF-α)、白介素-1β(IL-1β)、白介素-4(IL-4)和白介素-10(IL-10)的浓度。结果:(1)神蜂精治疗组均能显著降低大鼠右后足爪肿胀度(P<0.01);(2)神蜂精治疗组均能增加胸腺指数(P<0.05),降低脾脏指数(P<0.05);(3)神蜂精治疗组均能显著降低致炎性细胞因子TNF-α、IL-1β水平(P<0.01);(4)神蜂精高剂量组能显著增加抗炎性细胞因子IL-4、IL-10水平(P<0.01)。结论:神蜂精对AA大鼠关节炎症有一定的治疗作用,其可能机制是:保护胸腺和脾脏免疫器官,提高机体抗炎免疫能力;抑制致炎性细胞因子TNF-α和IL-1β的生成,促进抗炎性细胞因子IL-4和IL-10的产生,产生免疫调节作用。

摘  要:
Objective:To study the effects of Shen Feng Jing(SFJ)in adjuvant arthritic rats.Methods:Complete Freund's adjuvant(CFA)was injected to induce experimental arthritis in rats.60 SD male rats were randomly divided into the normal group,model group,SFJ high-dose group,SFJ middle-dose group,SFJ low-dose group,Compound Dexamethaso(CD)group with 10 rats in each.A model of experimental adjuvant-induced arthritic rats(AA rats) was established in the latter five groups.After 14 days,the six groups were treated with normal saline,1.0 mL/kg SFJ and 0.5,0.25 mL/kg SFJ,1.0g/kg CD by local treatment for 28 days.Paw swelling degree of AA rats was measured,and the immune organ weight was determined.The levels of TNF-α,IL-1β,IL-4 and IL-10 in serum were estimated by Enzyme-linked Immunosorbent Assay(ELISA)kits.Results:(1)All SFJ-treated groups decreased significantly the paw swelling degree(P<0.01);(2)All SFJ-treated groups increased the thymus index(P<0.05),reduced spleen index(P<0.05).(3)All SFJ-treated groups significantly decreased the levels of TNF-α and IL-1β;(4)SFJ high-dose group increased the content of IL-4 and IL-10 in the AA rats serum(P<0.01).Conclusions:SFJ could inhibit the inflammation of the AA rats.Mechanisms of therapeutic effects may be protecting immune organs,reducing the expression of TNF-α,IL-1β,and increasing the expression of IL-4,IL-10.
