贵州绵羊线粒体DNA(mtDNA)多态性研究

摘 要:
采用15种限制性内切酶对贵州威宁绵羊和六盘水绵羊各5只个体的mtD-NA多态性进行研究。其中只有BamHⅠ一种酶的酶切类型表现多态,产生三种酶切类型。共得到17种限制性态型,可归结为三种mtDNA单倍型,在威宁绵羊中单倍型Ⅰ所占比较较大(80%),单倍型Ⅲ只占20%,而在六盘水绵羊中则单倍型Ⅰ和Ⅱ所占比例相当;根据单倍型在群体中的分布和单倍型间遗传距离,推测贵州绵羊的mtDNA单倍型Ⅰ和Ⅱ可能代表两个母系祖先的基本单倍型,其分化时间约有16~32万年之久,单倍型Ⅲ则可能是由单倍型Ⅰ随机突变而来;两个地方绵羊群体间的分化时间大约为6.3~12.6万年,群体间的mtDNA遗传多态度较低,但六盘水绵羊群体内遗传多态度比威宁绵羊较丰富。

摘 要:
In this study 15 restriction enzymes were employed to investigate the mitochondrial DNA polymorphism among the 10 individuals from 2 local Guizhou sheep populations.Only one enzyme (BamH Ⅰ)was found to be polymorphous,which expressed three mtDNA restriction morphs.Seventeen restriction patterns were found that they could be sorted into 3 mtDNA haplotypes.The proportion of haplotype Ⅰ in Weining sheep is higher(80%),but that of haplotype Ⅲ is lower(20%),the proportion of haplotype Ⅰ and Ⅱ are almost equal among Liupanshui sheep.According to the proportion of haplotypes in the population and the genetic distances between the haplotypes,it is deduced that the mtDNA haplotype Ⅰ and Ⅱ are probably representing two mother ancestors of Guizhou sheep,which might separate around 160~320 thousand years ago,haplotype Ⅲ is probably coming from haplotype Ⅰ by random mutation;Two local Guizhou sheep populations separated about 63~126 thousand years ago,and the heterogeneity among the population is low,of which Liupanshui sheep is higher than Weining sheep.
