奶牛脐带间充质干细胞的体外培养与鉴定

摘  要:
【目的】在无菌环境下截取新生牛脐带20 cm,在体外进行原代奶牛脐带间充质干细胞分离培养,建立原代奶牛脐带间充质干细胞分离、培养、传代的方法,并进行形态学观察和多向分化能力检测。【方法】采用多重酶混合消化法,分离出原代奶牛脐带间充质干细胞,通过倒置显微镜下观察细胞形态,绘制细胞生长曲线,向成脂、成骨诱导分化。【结果】培养出了奶牛脐带间充质干细胞。倒置显微镜下观察,细胞形态呈相对均一的梭形,平行排列生长或呈旋涡状生长。能够诱导分化为成脂、成骨细胞。细胞增殖能力强,可多次传代。【结论】胰酶消化法能够快速分离出原代奶牛脐带间充质干细胞,所获得的细胞纯度较高,具有一般间充质干细胞的生物学特性,具有多向分化潜能。

摘  要:
【Objective】20cm of newborn calf umbilical cord in a sterile environment was intercepted and isolated in vitro,and then the cow umbilical cord mesenchymal stem cells were cultivated. This aimed to establish the method of the isolation,cultivation and passage of cows umbilical cord mesenchymal stem cells.And the morphological observation of them was carried out and their ability of multi- directional differentiation was detected.【Method】Multiple enzyme mixed digestion was used,primary cows umbilical cord mesenchymal stem cells were isolated and the cell morphology was observed by an inverted microscope,and the cells were induced into osteoblasts and adipocytes.【Result】Cows umbilical cord mesenchymal stem cells were successfully nurtured. Under an inverted microscope,cell morphology was relatively homogeneous spindle parallel growth or the growth was swirling. They can be induced into adipocytes and osteoblasts. The UC- MSCs have the ability of proliferation and can be passaged repeatedly. 【Conclusion】The trypsin digestion method can quickly separate the cows primary umbilical cord mesenchymal stem cells,and the obtained cells have high purity with the general biological characteristics of mesenchymal stem cells,and also have potentiality of multi- directional differentiation.
