褪黑激素定量生物检测方法的初步研究

摘  要:
用林蛙蝌蚪作实验动物,研究光照和褪黑激素(MT)浓度的变化对蝌蚪头部三角区、尾肌基部、尾肌基部(尾基)尾上鳍、尾肌中部、尾中部尾上鳍和尾尖黑色素颗粒聚集程度的影响,探索利用蝌蚪进行MT定量生物检测的条件。结果表明:控光(黑暗)条件下,头部三角区、尾肌基部、尾基尾上鳍色素细胞色素颗粒较自然光照组聚集程度明显(P<0.05),而尾尖不明显(P>0.05);在相同浓度的MT溶液中,头部三角区处理时间越长,其色素颗粒聚集程度越大(P<0.05),而尾中部尾上鳍不明显(P>0.05);在相同的实验条件下,在0.1~1.0ng/mL浓度范围内,MT浓度与蝌蚪色素细胞黑色素颗粒聚集程度相关,且下午时外源性MT对蝌蚪影响较大。因此,用林蛙蝌蚪作MT定量生物检测方法的模型动物是可行的,且其头部三角区和尾肌基部是较佳的观察、比较部位。

摘  要:
Using tadpoles of Rana chensinensis as experimental animal, we detected the effects of illumination and concentration of melatonin on tadpole melanin granule clustering degree, at head trigone, basal of tail muscle, tail basal ogami fins, cental of tail muscle, tail central ogami fins and tail tip, in order to explore the term using tadpoles to test melatonin at quantitative. The results showed that in darkness condition, the melanin granule in head trigone, basal of tail muscle and tail basal ogami fins had distinct collection (P < 0.05), but not at the tail tip (P > 0.05). Furthermore, head trigone had smaller melanin granule(P < 0.05), but not at the tail central ogami fins (P > 0.05) when treated with the same concentration MT solution. More importantly, it was found that the aggregating degrees of melanin granules of dermal melanophore were related to the melatonin concentrations within certain concentration range (0.1~1.0 ng/mL) in the same experimental condition and treatment in the afternoon had great effect. Therefore, using tadpoles of Rana chensinensis as experimental animal was available, and the head trigone and basal of tail muscle were better for examination and comparation.
