摘  要:
为研究开发具有应用价值的新型微生物资源,用邻硝基苯酚β-D-半乳糖苷(ONPG)法,从雪莲菌发酵乳中分离到一株产乳糖酶的酵母菌株XL-B36,通过分子鉴定,确定该菌为马克斯克鲁维酵母(Kluyveromyces marxianus)。对该菌所产乳糖酶酶学性能进行研究,该酶最适反应温度和最适pH值分别为47℃和7. 0,在35~40℃具有良好的稳定性。pH 5. 0~8. 0表现稳定,pH 7. 0时出现酶活峰值。金属离子Mn2+、Mg2+、Na+对酶活具有激活作用,Ca2+和Zn2+对酶活具有抑制作用,Cu2+和EDTA对酶活具有完全抑制作用。

摘  要:
In order to research and develop new microbial resources with applied value,a novel lactase-producing yeast XL-B36 was selected from fermented milk by ONPG method and identified as Kluyveromyces marxianus by molecular identification. The enzymatic properties of lactase produced by it were studied and the results showed that the temperature and pH value of β-galactosidase were 47℃ and 7. 0,respectively. The enzyme was stable at pH 5. 0 ~ 8. 0,and 35 ~ 40℃. The activity of β-galactosidase was promoted by Mn2 +,Mg2 +and Na+,but inhibited by Ca2 +and Zn2 +. It could be completely inhibited by Cu2 +and ethylene diamine tetraacetic acid(EDTA).
