两种大鼠心肌梗死模型的比较研究

摘 要:
对大鼠冠状动脉结扎法和药物注射法引起心肌梗死模型进行对比研究,探讨两种模型的优缺点。80只SD大鼠,随机分成冠状动脉结扎组和药物注射组。冠状动脉结扎组大鼠行气管插管连通动物呼吸机,打开胸廓暴露心脏,用6-0缝合线结扎大鼠左冠状动脉前降支;药物注射组按10 mg/100 g皮下注射异丙肾上腺素,连续注射2 d。两组模型分别于处理后24 h、48 h7、2 h、1周、2周处死动物,做心肌TTC染色、HE染色及IHC进行病理组织学观察、分析。TTC染色显示,二者均有灰黄色心肌梗死区域的出现,冠脉结扎组心肌梗死范围较大;而药物注射组出现的梗死范围较小、散在;HE染色显示,两组心肌组织均出现凝固性梗死,炎细胞浸润,继而有肉芽组织长入,最后形成疤痕。不同的是,结扎组心肌梗死较彻底,梗死面积的大小与结扎部位的高低呈正相关;药物组的梗死区域内偶尔可见有未坏死心肌,梗死灶主要分布在左心室壁和室间隔上,尤以靠近心尖部为重。IHC显示,两组均有心肌坏死凋亡相关基因p53和Caspase-3的强阳性表达。冠状动脉结扎组较药物注射组动物的死亡数量多,在操作方法上药物注射法较冠状动脉结扎法简便易行,容易操作掌握,不需要太多的仪器设备。二者均可有效复制心肌梗死模型,可用于对心肌梗死的研究,但各有其自身的优势和特点,科研工作可根据要求及实际需要合理选用。

Comparison of the myocardial infarction models were produced by the coronary artery ligation method and drug injection method were used to explore the advantages and disadvantages and applications of the two models.The 80 SD rats were randomly assigned to the coronary artery ligation groups and drug injection groups.The rats in coronary artery ligation group were endotracheal intubation and were connected with small animal ventilator,and then the heart was exposed,and the left anterior descending coronary artery in the upper 1/3 was ligated.The rats in drug injection group injected isoproterenol and continuous injection of two days.After two groups of rats were treated,the rats were killed respectively in 24h,48h,72h,1 w,2 w,and made heart TTC staining,HE staining and IHC for histopathological observation and analysis.The TTC staining showed:The rat models of myocardial infarction made by the coronary artery ligation and drug injection group appeared sallow region of myocardial infarction.The region of myocardial infarction in the coronary artery ligation groups mostly showed larger infarct range.The region of myocardial infarction in the drug injection groups mostly showed more smaller range and scattered.The HE staining showed:The myocardial tissue of two kinds of methods rats all emerged coagulation necrosis,inflammatory cell invasion,then,there was granulation tissue in myocardial infarction,the scar formea finally.In the coronary ligation groups,the myocardial cells mostly were complete infarction,and the size of infarct area was positively correlated with the degree of ligation site.The myocardial cells in the drug injection groups occasionally seen remnants of myocardial cells within the infarcted area.The infarct area of drug injection groups mainly distributed to the left ventricular wall and septum,especially near the heart apexelse.The IHC showed:Both two groups have a strong expression to cardiac apoptosis-related gene P53 and Caspase-3.The number of death in coronary artery ligation rats was more than that of drug injection groups and compared with the coronary artery ligation method,the drug injection method was easier to master,without a lot of instruments and equipment.The coronary artery ligation method and drug injection method were both effective to copy the model of myocardial infarction,and had their own advantages and characteristics.We can reasonably use of two methods based on technical requirements and the practical needs in our scientific research work.
